摘要
将嗜水气单胞菌 ( Ah) J-1株的 HEC毒素和胞外蛋白酶 ( ECP)分别与迟缓爱德华氏菌 ( Et) 3 8株的脂多糖 ( LPS)及脱毒多糖 ( PS)偶联 ,制成 3种亚单位二联疫苗 :HEC-PS、ECP-PS、HEC-LPS。将它们与 Ah J-1和 Et3 8全菌灭活疫苗一同分别免疫小鼠 ,并设注射 PBS对照组 ,检测各组小鼠体液免疫及细胞免疫水平。结果显示 ,用间接 ELISA、溶血抑制和全菌凝集试验检出这 3种亚单位疫苗的抗体水平无明显差异 ,并且都对 Et3 8有凝集活性。用 MTT法检测脾淋巴细胞转化率和腹腔巨噬细胞吞噬活性 ,显示亚单位疫苗的细胞免疫指标均明显高于对照组 ,但略低于全菌苗免疫组。在第 2次免疫后的第 6周 ,分别用 1 0 0 LD5 0 Ah J-1和2 .3 7× 1 0 7CFU Et3 8攻击 ,Ah J-1攻击各组的保护率依次为 ,HEC-PS组 83 .3 %、ECP-PS组 80 %、HEC-LPS组 92 .3 %、Ah J-1全菌组 1 0 0 %、Et 3 8全菌组 2 5%、对照组无保护力 ;Et 3 8攻击各组的保护率依次为HEC-PS组 90 %、ECP-PS组 90 .9%、HEC-LPS组 85.7%、Ah J-1全菌组 62 .5%、Et3 8全菌组 1 0 0 %、对照组为 1 2 .5%。表明
With the extracellular proteins ( HEC toxin or ECP ) of Aeromonas hydrophila(Ah) and the lipopolysaccharide (LPS) or polysaccharide (PS) of Edwardsiella tarda( Et ), three conjugated subunit vaccines (CSV) were prepared, designated as HEC PS, ECP PS, HEC LPS. All mice for test were divided into 6 groups, which were immunized with the above three CSVs, inactiviated whole bacteria vaccine (WBC) of Ah J 1 or Et 38 strain, and PBS respectively. The titer of antibodies was tested with indirect ELISA, haemolytic invalid (HLI) test and whole bacteria agglutination (WBA) test. The results showed that there were no significant differences between the above three CSVs and the two WBCs. At the 7th day after the second immunization, the stimulation index (SI) of the spleen lymphocyte transformation (SLT) and the A 570 of abdomen macrophagocyte phagic activity (AMA) were measured with MTT method. The results showed that as far as the values of the SI and AMA of the mice, WBCs were better than CSVs, and the control mice had the lowest value. At the 6th week after the second immunization, all were challenged with 100 LD 50 Ah J 1 or 2.37×10 7 CFU Et38 per mouse separately. 83.3% of the mice immunized with HEC PS, 80% with ECP PS, 92.3% with HEC LPS, 100% with Ah J 1, 12.5% with Et 38, none with PBS was protected against Ah J 1 attack; while to the Et 38 challenge, the protect ratios were 90%, 90.9%, 85.7%, 62.5%, 100%, 12.5% accordingly. It suggested that all of the three CSVs could be used as combined vaccines against the challenge of Ah and Et. [WT5HZ]
