摘要
用致病性嗜水气单胞菌(Aeromonashydrophila,Ah)J1株制得全菌苗和毒素苗;从草鱼中制得α2巨球蛋白(α2M)。将150尾剑尾鱼分成5组,每组30尾。4个试验组分别用含有全菌苗、毒素苗、草鱼α2M+全菌苗、α2M+毒素苗的普通饲料连续投喂15d后,再投喂普通饲料20d;对照组用普通饲料连续投喂35d。用AhJ1株50LD50腹腔注射攻击。实验结果表现,全菌苗组剑尾鱼的相对保护率为43.3%,全菌苗+α2M组的相对保护率为53.3%,毒素苗组的相对保护率为26.7%,毒素苗+α2M组的相对保护率为30%,对照组全部死亡。χ2检验结果表明,各免疫组与对照组差异极显著,添加与不添加α2M的各组差异不显著,但添加α2M能延缓剑尾鱼的死亡时间。
Aeromonas hydrophila J-1 strain was cultured in LB medium,at 28 ℃ for 24 hours.After centrifugation and inactivated the whole bacteria and the toxin vaccine were prepared.α2-macroglobulin(α2M) were purified from grass carp plasma.The feeds were manufactured containing the whole bacteria,the toxin,the whole bacterium with α2M,and the toxin with α2M respectively,as well as the common feed as the control.150 Xiphophorus helleri were divided into five groups,with 30 fish per group.The above feeds were fed to experiment fishes respectively for successive 15 days and common feed for further successive 20 days.All five groups were challeged with AhJ-1 strain 50 LD 50. The whole bacteria vaccine had the relative percentage survival(RPS)of 43.3%,the whole bacteria vaccine with α2M 53.3%,the toxin vaccine 26.7%,the toxin vaccine with α2M 30%,while the negative control fishes died completely.χ2 test showed that the difference between the immunized groups and the control group was significantly.The difference between the groups with or without α2M was not obviously but α2M could delay the death time of Xiphophorus helleri.
