蝮蛇毒抗高凝状态酶对小鼠移植肝癌作用及骨髓保护的实验研究

摘要 目的建立实验性小鼠肝癌原位移植模型,在建立模型基础上研究蝮蛇毒抗高凝状态酶(AHCSE)的抑癌作用及骨髓保护作用。方法用小鼠肝癌H22细胞种植于小鼠腋窝皮下,传代后取癌组织块种植于小鼠肝脏,进行蛇毒制剂治疗的探索。将带有肝癌H22小鼠分成四组:A组(NS组)、B组[阿霉素(ADM)组]C组(AHCSE组)、D组[阿霉素(ADM)+AHCSE组],每组10只。给药10 d后,根据瘤重、瘤体积计算抑瘤率;观察骨髓有核细胞数。结果不同治疗组实验表明,AHCSE组、ADM组和联合组瘤重及瘤体积抑制率分别为(64.25%、68.58%)(、70.14%7、2.77%)和(82.08%、85.39%),且AHCSE在化疗过程中有刺激骨髓增生作用,可保护骨髓组织,拮抗化疗的毒副作用。结论 AHCSE对小鼠原位移植肝癌H22的生长具有抑制作用,且对小鼠骨髓具有一定的保护作用。 Objective To explore the antitumor and bone marrow protective effect of anti-hypercoagulability state enzyme (AHCSE) from agkistrodon venom by establishing experimental mouse model bearing hepatoma 14222 strain in situ. Methods Hepatoma H22 cells were planted in mouse subcutaneous axillary, and then the cancer tissue were planted in mouse liver after passage. Finally, the treatment effect of venom preparations was explored. Mice bearing H22 were divided into 4 groups : group A ( NS), group B ( adriamycin), group C (AHCSE), group D (adriamycin + AHCSE). Each group contained 10 mice. For ten days' drug delivery, the tumor inhibition rate was calculated according to the tumor weight and volume. The nucleated cells in bone marrow were counted. Results The tumor weight and volume inhibitory rates in different treatment group (AHCSE, ADM and combination group) were (64. 25%, 68. 58% ), (70.14% ,72.77% ) and (82.08% ,85.39% ) respectively and the AHCSE might stimulate bone marrow hyperplasia, protected bone marrow tissue and antagonized the side effect of chemotherapy during the course of treatment. Conclusion The AHCSE may inhibit the growth of hepatoma H22 strain in mice and has a protective effect on bone marrow.
