烤烟和白肋烟的致突变作用和细胞毒性的对比研究 被引量:2

摘要 目的比较纯粹烤烟和白肋烟的致突变作用和细胞毒性,为开发低危害卷烟提供参考依据。方法以2种纯粹烤烟(F1,F2)和2种纯粹白肋烟(B1,B2)为研究对象,应用鼠伤寒沙门杆菌TA98和TA100菌株,分别在代谢活化和非活化条件下进行细菌回复突变试验;应用人淋巴母细胞TK6细胞进行TK基因突变试验,并通过测定相对悬浮生长率和平板效率考察细胞毒性,应用中国仓鼠成纤维细胞(CHL)细胞进行中性红试验。结果细菌回复突变试验结果表明,在非代谢活化条件下,4个样品均未检测出致突变作用;在代谢活化条件下,只有白肋烟样品被检测出具有较弱致突变作用。TK基因突变试验结果显示,与阴性对照组相比,白肋烟样品(B2)显著诱导突变频率的增加,差异具有统计学意义。TK6细胞细胞毒性试验和CHL细胞中性红试验结果表明,未见烤烟和白肋烟的细胞毒性之间差异有统计学意义。结论在该实验条件下,认为白肋烟的致突变作用强于烤烟。 [Objective]To compare the mutagenicity and cytotoxicity between pure flue-cured tobacco and burley tobacco,and provide reference basis for developing less harmful cigarette.[Methods]Two pure flue-cured tobacco(F1,F2) and two burley tobacco(B1,B2) were taken as study objects.The mutagenicity of the samples was evaluated by bacterial reverse mutation test in Salmonella TA 98 and TA100 under metabolic activation and non-activated conditions.TK gene mutation assay was conducted in TK6 cell.The cytotoxicity of the samples was evaluated through determination of relative suspension growth and flat panel efficiency.CHL cell was used for neutral red test.[Results] The bacterial reverse mutation test showed that the four samples had no mutagenicity without metabolic activation,only the burley tobacco samples had a weak mutagenicity with metabolic activation.The TK gene mutation assay revealed there was a statistically significant increase in mutant frequency of B2 that compared with the negative control.Both TK gene mutation assay and neutral red uptake assay showed no difference in cytotoxicity between flue-cured samples and burley samples.[Conclusion]The mutagenicity of burley tobacco is stronger than flue-cured tobacco in this test condition.
