摘要
目的探讨雷公藤内酯对海人酸致痫大鼠海马神经元的保护作用及对海马神经元PUMA(P53上调的细胞凋亡调控因子)蛋白表达的影响。方法 30只SD大鼠随机分成3组,其中对照组10只,海人酸组10只,雷公藤内酯干预组10只。结晶紫染色观察海马神经元的形态变化,免疫组化染色法检测海马神经元PUMA蛋白的表达。结果结晶紫染色结果显示,海人酸组中,海马神经元变性或丢失,胞体皱缩,形态不规则。而雷公藤内酯干预组中海马神经元的数量和形态与对照组相似,胞浆清晰淡染,核仁清楚。免疫组化染色结果显示,海人酸组中海马神经元PUMA蛋白表达水平与对照组比较显著增多(P<0.05);雷公藤内酯干预组中海马神经元PUMA蛋白表达水平与海人酸组比较显著减少(P<0.05)。结论雷公藤内酯对海人酸致痫大鼠海马神经元具有保护作用;其可下调海马神经元PUMA蛋白的表达,抑制海人酸致痫大鼠海马神经元凋亡。
Objective To investigate the effect of triptolide on the hippocampal neuron in kainite-induced rat and the expression of PUMA protein of the hippocampal region. Methods 30 SD rats were divided into 3 groups, per group 10 rats, including control group, kainite-induced group and triptolide treatment group. Crystal violet staining was used to observe the morphological change of the hippocampal neuron. Immunohistochemistry staining was used to examine the expression of PUMA protein of the hippocampal region. Results Crystal violet staining showed that the neurons of the hippocampal region exhibited irregular morphology and body shrinked, and rednced in numbers in kainite-induced group. However, the neurons of triptolide treatment group showed clear cytoplasm dyed and nucleoli, and had the similar number and morphology compared with the control group. Immunohistochemistry staining showed that the expressions of PUMA on neurons in hippocampal region in kainite-induced group were increased significantly than in control group(P 〈0.05 ). The expressions of PUMA on neurons in hippocampal region in triptolide treatment group were obviously decreased than kainite-induced group ( P 〈 0.05 ). Conclusion Triptolide can be of protective effect on the hippocampal neurons in kainite-induced rat, and can inhibit the apoptosis of neurons in the hippocampal region through deregulating the expression of PUMA protein.
