和解聚散方对单侧输尿管梗阻大鼠CTGF和LN表达的影响 被引量:6

Effect of Hejie Jusan Formula on Expressions of CTGF and LN of Rats with Unilateral Ureteral Obstruction

摘要 目的:观察和解聚散方对单侧输尿管梗阻(UUO)致大鼠肾间质纤维化的结缔组织生长因子(CTGF)和层黏连蛋白(LN)的影响。方法:采用UUO建立大鼠肾间质纤维化模型,随机分为模型组、贝那普利组和和解聚散方组,给予相应药物灌胃,并设假手术组;UUO术后7天、14天和21天取材,采用免疫组化法检测各组手术侧肾组织CTGF和LN的表达。结果:与假手术组相比,UUO模型组7天、14天和21天的CTGF和LN的表达均增高(P<0.05);与模型组相比,和解聚散方组7天、14天和21天肾组织中的CTGF和LN表达均减少(P<0.05);21天和解聚散方组CTGF的表达比贝那普利组显著降低(P<0.05)。结论:和解聚散方可以降低肾间质纤维化不同时间点CTGF和LN的表达,表明了和解聚散方的有效性,验证了肾络聚散消长病机学说指导下的和解聚散法为延缓慢性肾脏病肾间质纤维化进展的有效治法。 Objective: To study the effect of Hejie Jusan formula on expressions of connective tissue growth factor(CTGF)and laminin(LN)in unilateral ureteral obstruction(UUO)-induced renal interstitial fibrosis(RIF)rats.Methods:RIF model rats were established by UUO.All the UUO rats were randomly divided into model group,benazepril group and Hejie Jusan formula group.Healthy rats were chosen as the sham operation group.The rats were killed on the seventh,fourteenth and twenty-first day in batches respectively after UUO.The protein expressions of CTGF and LN in kidney tissues were detected by Immunohistochemistry.Results:Compared with sham operation group,the protein expressions of CTGF and LN in model groups of three time points were all significantly increased(P0.05).Compared with model group,the protein expressions of CTGF and LN in Hejie Jusan formula group at three time points were all significantly decreased(P0.05).And the expression of CTGF on 21st day of Hejie Jusan formula group was significantly decreased compared with that of benazepril group(P0.05).Conclusion:Hejie Jusan formula has the function of down-regulating the expressions of CTGF and LN in RIF rats at three time points after UUO,which suggests the effectiveness of Hejie Jusan formula,proves Hejie Jusan method is the effective method to delay the progress of RIF in chronic kidney disease under the theory of kidney collaterals.
