摘要
目的:观察大黄虫丸对免疫性肝纤维化大鼠血流变和微循环变化的影响,从"络病"角度探讨其抗肝纤维化的中医病机。方法:制备猪血清致大鼠肝纤维化模型,观察大黄虫丸对该模型动物的全血黏度150/s切变率、30/s切变率、5/s切变率、1/s切变率、红细胞压积、舌下及耳廓的脉络变化的影响。结果:大黄虫丸能显著降低肝纤维化大鼠的全血黏度150/s切变率、30/s切变率、5/s切变率、1/s切变率、红细胞压积、透明质酸酶(HA)、层黏连蛋白(LN)、Ⅲ型前胶原(PCⅢ)、IV型胶原(CIV)、谷丙转氨酶(ALT)、谷草转氨酶(AST)指数,改善纤维化大鼠舌下和耳廓脉络的循环状态。结论:大黄虫丸对免疫性肝纤维化大鼠血流变和微循环变化的改善,提示"活血通络"可能是其抗肝纤维的主要病机之一。
Objective:To observe effects of Dahuang Zhechong Pill on hemorheology and microcirculation in rats with immunological hepatic fibrosis and attempt to use the theory of collateral disease to explain the mechanism of Dahuang Zhechong Pill's resistive effects on hepatic fibrosis.Methods:Rat fibrosis models were established by pig serum.Hematocrit and blood viscosity at shear rate in 150/s,30/s,5/s,1/s were detected respectively among control,model and Dahuang Zhechong Pill groups;at same time,microcirculation microscope was used to observe the thread of hypoglottis and auricle in rats.Results:Dahuang Zhechong Pill could reduce the indexes of hematocrit,blood viscosity at different shear rate,Hyaluronic Acid(HA),Laminin(LN),Procolagen Type Ⅲ(PCⅢ),Collagen TypeⅣ(CⅣ),glutamic-pyruvic transaminase(ALT)and glutamic-oxalacetic transaminase(AST)in hepatic fibrosis,and improve the microcirculation of hypoglottis and auricle in model rats.Conclusion:The improvement of blood rheology and microcirculation suggest the mechanism of Dahuang Zhechong Pill's resistive effects on hepatic fibrosis is associated with promoting blood circulation to remove meridian obstruction.
