摘要
目的探讨淫羊藿总黄酮对高脂血症大鼠体内过氧化物酶体增殖物激活受体(PPARγ)表达水平和氧化应激水平的影响。方法将健康SD雄性大鼠35只随机分为普通饲料组9只和高脂饮食组26只。高脂饮食组给予高脂饲料喂养12周后复制高脂血症大鼠模型。造模成功后随机分为模型对照组8只、淫羊藿大剂量组(200 mg/d)9只及淫羊藿小剂量组(100 mg/d)9只,干预4周后观察各组大鼠血脂水平,检测各组大鼠的超氧化物歧化酶(SOD)活性、血清丙二醛(MDA)水平及PPARγ的表达水平。结果模型对照组甘油三酯(TG)、总胆固醇(TC)水平均明显高于普通饲料组(P<0.01)。淫羊藿总黄酮大、小剂量组TG、TC水平均明显低于模型对照组(P<0.05,P<0.01),且淫羊藿总黄酮大剂量组对TG、TC水平的改善较淫羊藿总黄酮小剂量组更为明显(P<0.05)。淫羊藿总黄酮大、小剂量组对高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)水平的影响无统计学意义(P>0.05)。模型对照组大鼠干预后SOD活性较普通饲料组降低(P<0.01),MDA水平升高(P<0.01)。淫羊藿总黄酮大、小剂量组SOD活性均较模型对照组升高(P<0.01),MDA水平降低(P<0.01),且淫羊藿总黄酮大剂量组较淫羊藿总黄酮小剂量组更能明显增加SOD活性(P<0.01)和降低MDA水平(P<0.01)。模型对照组大鼠干预后PPARγ表达水平较普通饲料组明显降低(P<0.05)。淫羊藿总黄酮大、小剂量组大鼠PPARγ表达水平均较模型对照组明显升高(P<0.01),且淫羊藿总黄酮大剂量组升高明显(P<0.01)。结论淫羊藿总黄酮能够提高高脂血症大鼠血清SOD水平,降低MDA水平,改善氧化应激,对内皮有保护作用,其机制可能是上调体内PPARγ的表达水平。
Objective To investigate the influence of total flavonoids of epimedium (TFE) on the oxidative stress and the expression of peroxisome proliferators activated receptor gamma in hyperlipemia rats. Methods 35 male SD rats were randomly divided into the high -fat diet group(26) and full diet group(9). High -fat diet group were fed for 12 weeks ,then 26 male SD rats were randomly divided into three groups: the hyperlipemia group (8), group of high hyperlipemia with a large dose of TFE (9,200 rag/d) and group of high hyperlipemia with a small dose (9, 100 mg/d). After 4 weeks of intervention with TFE, the activation of superoxide dismutase, the level of serum malonaldehyde and the expression of peroxisome proliferators activated receptor gamma was measured. Results The levels of TG, TC and LDL in hyperlipemic group were significantly increased compared with fun diet group after 12 weeks ( P 〈 O. 01 ), which indicates that the model of hyperlipemia has been established. Compared with the con- trois, the activation of superoxide dismutase was elevated, and the level of serum malonaldehyde was decreased while the expression of peroxisome proliferators activated receptor gamma was attenuated after the intervention with TFE. Conclusion TFE could elevate the activation of superoxide dismutase, decrease the level of serum malonaldehyde and elevate the expression of peroxisome proliferators activated receptor gamma in hyperlipemia rats, which means it may play an important role in protect the function of endo- thelial cells from oxidative injury via up - regulating the expression of nuclear peroxisome proliferators activated receptor gammma.
