摘要
目的探讨金银花水提取总黄酮(water extraction from honeysuckle total flavonoids,WEHF)对高血脂模型小鼠的降血脂作用及机制。方法应用高脂饲料建立高血脂模型小鼠的基础上,将50只小鼠随机分为正常对照组(10只)和造模组(40只)。造模组每天饲喂高脂饲料,正常组每天饲喂正常饲料,连续灌喂WEHF 28d。检测各组小鼠的体质量、饮食量、饮水量、血脂常数、肝脏指数、肾脏指数;检测肝脏内过氧化氢酶(catalase,CAT)活性、超氧化物歧化酶(superoxide dismutase,SOD)活性以及丙二醛(malondialdehyde,MDA)含量;检测胰岛素受体底物1(insulin receptor substrate,IRS1)、低密度脂蛋白受体(low density lipid receptor,LDLR)、载脂蛋白1(apolipoprotein 1,Apo-1)、脂肪酸合成酶(fatty acid synthase,FAS)、固醇调节元件结合蛋白(sterol regulatory element binding protein-1c,SREBP-2)和6-磷酸果糖激酶-2(6-phosphofructo-2-kinase/fructose-2,6-biphosphatase 2,PFK-2)等mRNA表达;检测LDLR和SREBP-2的蛋白表达。结果模型组小鼠的体质量显著低于正常组(P<0.05);饮食量和饮水量显著高于正常组(P<0.05),且总胆固醇(Total cholesterol,TC)值为4.09mmoL/L,模型组与正常组小鼠的TC值有显著差异(P<0.05),说明造模成功。与正常组比较,模型组肝脏指数显著上升而肾脏指数显著下降(P<0.05)。与模型组相比,中、高剂量WEHF能显著降低高血糖小鼠血浆甘油三酯(Trilaurin,TG)水平(P<0.05);高剂量WEHF的能显著降低高血脂小鼠血浆TC水平(P<0.05);各WEHF组均能显著降低MDA含量(P<0.05)和显著提高SOD活性(P<0.05);WEHF中剂量、高剂量组能显著升高CAT水平(P<0.05);高剂量的WEHF能显著上调IRS1、LDLR、Apo-1、FAS、SREBP-2和PFK-2等mRNA表达水平(P<0.05);中、高剂量的WEHF能显著上调LDLR和SREBP-2的蛋白表达水平(P<0.05)。结论WEHF能改善高脂诱导小鼠肝脏的氧化应激水平,从而维持血液中正常脂质水平。其机制可能与WEHF上调模型小鼠肝脏IRS1、LDLR、Apo-1、FAS和SREBP-2等mRNA表达水平,上调LDLR以及SREBP-2转录因子的蛋白表达水平有关。
Objective To explore the effect and mechanism of water extraction from honeysuckle total flavonoids(WEHF)on blood lipid in hyperlipidemic mice.Methods Hyperlipidemic mice model was created by feeding mice with high-fat diet,and 40 hyperlipidemic mice were randomly divided into 4 groups and were orally given water(model control group),high dose WEHF(500 mg/kg),medium dose WEHF(300 mg/kg)and low dose WEHF(100 mg/kg)for 28 days,10 mice in each group.Meanwhile,10 wild type mice were included as healthy controls.The body weight,food and water intake,lipid parameter,liver and kidney index were determined.Catalase(CAT)activity,superoxide dismutase(SOD)activity and malondialdehyde(MDA)content in liver were measured.The mRNA levels of insulin receptor substrate 1(IRS1),low density lipoprotein receptor(LDLR),apolipoprotein 1(Apo-1),fatty acid synthase(FAS),srebp-2 monoclonal antibody2(SREBP-2),and 6-phosphofructokinase-2(PFK-2)as well as protein levels of LDLR and srebp-2 were compared.Results The differences in body weight,food and water consumption and serum TC level were significant between hyperlipidemic mice and healthy controls(P<0.05).In hyperlipidemic mice,the values of hepatic panel increased while the values of renal function panel decreased significantly(P<0.05).The serum levels of TG and TC were significantly lower in WEHF medium dose and high dose group than that in model control group(P<0.05).MDA content in liver decreased in all hyperlipidemic mice while CAT level increased in medium and high dose WEHF mice(P<0.05).The SOD activity in liver elevated in low,medium and high dose WEHF mice,and the mRNA levels of IRS1,LDLR,Apo-1,FAS,SREBP-2 and PFK-2 were significantly higher in high dose WEHF mice(P<0.05).The protein levels of LDLR and SREBP-2 were also higher in medium and high dose WEHF mice(P<0.05).Conclusions WEHF may help to maintain the normal level of blood lipid by enhancing the oxidative response,up-regulating the transcription of IRS1,LDLR,Apo-1,FAS,SREBP-2 and the expression of LDLRS and SREBP-2 in liver of high-fat induced hyperlipidemic mice.
