摘要 以甜叶菊茎段为外植体、MS为基本培养基,研究了消毒剂70%酒精和0.1%升汞的不同时间处理组合的灭菌效果和对试管苗再生的影响,同时还探讨了不同种类、不同浓度植物生长调节剂对试管苗增殖、生根及移栽的影响。结果表明7,0%酒精处理15~30s+0.1%升汞处理4m较为适合茎段外植体灭菌,污染率低、苗再生率高;6-BA与NAA组合的试管苗增殖系数比KT、Ad与NAA组合要高,培养基中添加1.0mg/L 6-BA+0.1~0.2mg/L NAA植物生长调节剂组合适合试管苗增殖,增殖系数可达7.4;培养基中添加0.0~0.5mg/L的IBA或NAA均适合试管苗生根,生根的试管苗移栽成活率较高。 The stem -sections were chosen as explants for plant tissue culture in the MS basic medium. The test investigated the influences of disinfectors 70% alcohol and 0.1% HgC12 on sterilizing effects and shoot regenera- tion at first. And then, this test studied the effects of the plant growth regulators on shoot proliferating, rooting and transplanting too. The results showed that 70% alcohol treated for 15 - 30s with 0.1% HgCI2 treated for 4m was suitable for explants sterilizing and got lower explants infectious ratio and higher shoot formation ratio. The combinations of 6 - BA and NAA got higher proliferation coefficient than the combinations of Ad and NAA or the combinations of KT and NAA. The basic MS medium with 1.0mg/L 6 - BA and 0.1 - 0.2mg/L NAA was suit- able for shoot proliferation, and the coefficient of proliferation could reach 7.4. The 1/2 MS medium with 0.0 - 0.5mg/L IBA or NAA was suitable for shoot rooting and the survival rate was high.
