亲和层析分离纯化乳酸脱氢酶

Separation and Purification of Lactate Dehydrogenase by Affinity Chromatography

摘要 以壳聚糖为载体,戊二醛为交联剂,氨基三乙酸为配基,制备固载铜离子的金属螯合亲和吸附剂,层析分离纯化乳酸脱氢酶(LDH),收得率为23.3%,纯化倍数达3.1。实验结果表明该亲和层析固定相对LDH有较高的分离纯化选择性。 With using chitosan as the support,glutaraldehyde as the crosslinking agent and nitrilotriacetic acid as the chelating ligand,affinity adsorbent was prepared by immobilized copper ion.Lactate dehydrogenase(LDH) was purified by chromatography with a recovery rate of 23.3% and purfication fold of the 3.1.The results show that the affinity chromatography has a high selectivity for the separation and purification of LDH.
