摘要
采用RT-PCR方法,扩增并测定了黄瓜绿斑驳花叶病毒广东分离物(CGMMV-GD-GZ,CGMMV-GD-LZ,CGMMV-GD-LF)的外壳蛋白(coat protein,CP)和运动蛋白(movement protein,MP)基因序列。结果显示,CP基因序列全长486 bp,编码161个氨基酸;MP基因全长795 bp,编码264个氨基酸。CGMMV广东分离物与其他15个CGMMV分离物之间外壳蛋白基因核苷酸的同源性在90.9%~100%之间,运动蛋白核苷酸同源性为92.7%~100%;与同属其他13种病毒基因组核苷酸序列同源性分别仅为29.5%~56.2%和38.8%~61.8%。基于外壳蛋白和运动蛋白基因序列构建系统发育树显示,CGMMV不同分离物进化为亚洲和欧洲两大群体,3种CGMMV广东分离物与韩国、日本等亚洲分离物同源性极高,可能具有共同的侵染源。
The coat protein and movement protein gene of Cucumber green mottle mosaic virus(CGMMV-GZ,CGMMV-LZ,CGMMV- GF) in Guangdong province was amp lifted by RT-PCR. The results revealed that the three isolates contains complete coat protein gene and movement protein gene, which were 486 and 795 nucleotides respectively, and encodes the coat protein of 161 amino acids and the movement protein of 264 amino acids. Multiple sequences aligment comparing to other CGMMV isolates which downloaded from GenBank showed that the nucleotide identities of coat protein are 90.9%-100%, while movement protein were 92.7%-100% among all the tested isolates, but only shared identities of 29.5%-56.2% and 38.8%-61.8% with other Tobamoviruses. Homology trees generated from movement protein and coat proteins suggested that CGMMV could be separated into two subgroups. There was no marked diversity between CGMMV- GD and other Asia isolates, they presented very close relatives among each other that from different countries, and might so far as to share the same infected source.
