摘要
目的研究适合基层法医DNA实验室建立DNA数据库的新产品和方法.方法采用国产Goldeneye 20A-M免提取试剂盒对3 977份多种建库人体生物检材进行检测,并对多种人体生物检材的结果进行比较.同时探索Whatman FTA卡的洗脱方法.结果直接用免提取试剂盒进行实验研究,检出率为99.4%,对保存了1 a的普通血滤纸、棉签血痕、纱布血痕、AuSBio卡、公安部物证鉴定中心监制的采血卡检出成功率高达100%,口腔拭子和FTA分别为97.1%和98.5%.Whatman FTA卡经过1%的Triton X-100洗脱1次,检出成功率可以达到100%.结论 Goldeneye 20A-M免提取试剂盒可以用多种载体人体生物检材做为直扩样本进行DNA建库,是一种简单、快捷的DNA建库方法.
Objective To search new product and new idea for establishing DNA database at the grassroots level.Methods We tested 3977 blood samples by direct amplification Kit Goldeneye 20A-M and compared the success rate amont many types of blood samples.Meanwhile,we also explored the elution process of Whatman FTA card.Results The total checkout success rate of all blood samples by Goldeneye 20A-M kit was 99.4%.The checkout success rate was 100% for common blood filter paper,cotton bud blood sample,bandage blood sample,AuSBio card and blood collecting card made by evidence centre of ministry of public security.The checkout success rate for buccal swab and FTA was 97.1% and 98.5%,respectively.FTA＇s checkout success rate was 100% after washing by 1% Triton X-100.Conclusion The direct amplification Kit Goldeneye 20A-M from many types of blood samples is effective for STR analysis,and is a quick and easy method for establishing DNA database.
