摘要
目的应用HPLC对自然突变品种红金银花中化学成分进行初步分析,建立了其中木犀草苷的分析方法并测定其含量。方法选用Venusil XBP-Cl8柱,以乙腈-磷酸-水为流动相进行梯度洗脱,采用Agilent DAD检测器(238 nm)对自然诱变品种红金银花中木犀草苷进行了初步分析测定。结果木犀草苷的保留时间为39.50 min,定量线性范围0.035 5~ 0.284 0μg(r=0.999 9),精密度和稳定性实验RSD均小于3%,加样回收率达98.85%。结论 建立的红金银花中有效成分木犀草苷检测方法灵敏度高、稳定性和重现性好;红金银花中木犀草苷平均含量为0.54 mg/g。
Objective To establish an HPLC method for the analysis of chemical constituent and determined the content of luteolin in variety of natural mutation-Lonicerajaponica var chinensis. Methods HPLC analysis was performed on Venusil XBP - C18 ( 250 mm×4.6 mm, 5 μm ) column with mobile phase of acetonitrilephosphoric acid-water and the detection wavelength was 238 nm. Results The retention time of luteolin was 39.50 min, the linear range was 0.035 5 - 0.284 0 μg ( r=0.999 9 ).The RSD of reproducibility and stability experiments was lower than 3% and the average recovery rate of luteolin is 98.85%.Conclusion This method was simple and fast with more accuracy and reproduciblity to detect luteolin in Lonicerajaponica var. chinensis. The average content of luteolin in Lonicerajaponica var. chinensis was 0.54mg/g.
