鸡骨草醇提物对大鼠非酒精性脂肪肝的保护作用研究 被引量:8

Protective Effect of Ethanol Extraction of Abrus cantoniensis on Non-alcoholic Fatty Liver in Rats

摘要 目的:研究鸡骨草醇提物对高脂饲料加四氯化碳(CCl4)所致大鼠脂肪肝的保护作用。方法:采用高脂饲料加四氯化碳(CCl4)复合诱导大鼠脂肪肝模型.实验分为5组,即正常对照(生理盐水2mL.kg-1)、模型(生理盐水2mL.kg-1)与鸡骨草醇提物高、中、低剂量(40、30、20g.kg-1)组,ig给药,每天1次,连续2周。检测大鼠血清中天冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)含量及体重、肝重,计算肝脏系数,并观察各组大鼠肝组织病理学变化。结果:与模型组比较,鸡骨草醇提物高、中、低剂量组大鼠血清中AST、ALT含量及肝脏系数显著降低,AST/ALT值升高(P<0.01);大鼠肝脏系数显著降低(P<0.05);肝组织病理学明显改善。结论:鸡骨草醇提物可降低病鼠肝脏系数和血清中ALT、AST含量,减轻肝细胞脂肪变性及病理损伤,对高脂饲料配合CCl4所致大鼠脂肪肝有改善作用。 OBJECTIVE:To study the protective effect of ethanol extraction of Abrus cantoniensis on fatty liver induced by CCl4 and high-fat feeding in rats.METHODS:Fatty liver model was induced by CCl4 and high-fat feeding.Model rats were divided into 5 groups,i.e.normal control group(normal saline 2 mL·kg-1),model group(normal saline 2 mL·kg-1) and ethanol extraction of A.cantoniensis high-dose,medium-dose and low-dose groups(40,30,20 g·kg-1).They were given relevant drugs intragastrically once a day for 2 weeks.The contents of AST and ALT,body weight and liver weight were determined and liver index were calculated.The histopathological changes of liver was also observed by microscope.RESULTS:Compared with model group,serum levels of ALT and AST and the liver index in ethanol extraction of A.cantoniensis high-dose,medium-dose and low-dose groups were decreased significantly while AST/ALT increased(P0.01),rat liver coefficient uere reduced(P0.05),liver tissue pathology were improved obviously.CONCLUSION:Ethanol extraction of A.cantoniensis can reduce the content of ALT and AST in serum and liver index,and relieve the pathologic damage and the degree of fatty degeneration of hepatic tissue.Ethanol extraction of A.cantoniensis can improve fatty liver induced by CCl4 and high-fat feeding in rats.
