摘要 基于亲水相互作用色谱(HILIC)技术,建立了可用于乳制品中尿素测定的液相色谱方法,其中固定相为Polar-Silica色谱柱,流动相为4mmol/L磷酸二氢铵溶液(pH=6.8)-乙腈(20:80,体积比),检测波长为200nm,流速为1mL/min。在该体系下,尿素保留时间适中,可与其他几种含氮极性化合物如三聚氰胺、三聚氰酸、三聚氰酸一酰胺、三聚氰酸二酰胺等实现基线分离。应用该方法测定了奶粉和液态奶制品中尿素的质量分数,其中奶粉中尿素的质量分数为1600~2400mg/kg,液态奶制品中尿素的质量分数为300~400mg/kg。 Urea in milk powder and liquid products was detected by using hydrophilic interaction liquid chromatography (HILIC). The sepa ration was carried out by using a Polar-Silica column as stationary phase and 4 mrnol/L NH4H2PO4(pH=6.8)-acetonitrile (20:80, v/v) as mobile phase. The detection was set at 200 nm and the flow rate was lmL/rnin. It was found that urea had moderate retention time on the Polar-Silica column. Baseline separation could be achieved between urea and melamine as well as its derivatives such as cyanuric acid, ammelide and ammeline. By using this method, the content of urea in milk powder and liquid products that were purchased from local supermarkets was determined. It was shown that the content of urea in the milk powder products was 1 600-2 400 mg/kg and that in the milk liquid products was 300-400 mg/kg.
