糖皮质激素诱导的亮氨酸拉链蛋白与多发性骨髓瘤骨病发展的关系 被引量:1

Investigation of the relationship between glucocorticoid induced leucine zipper and bone lesions of multiple myeloma

摘要 目的:研究糖皮质激素诱发的亮氨酸拉链蛋白(glucocorticoid induced leucine zipper,GILZ)在多发性骨髓瘤(MM)骨病发展中的作用。方法:收集MM患者抗凝血11例,利用密度梯度离心法获取外周血单个核细胞(PBMCs),并将PBMCs分为地塞米松(DEX)干预组及空白对照组,相同条件培养48h。采用逆转录-酶联聚合反应(RT-PCR)法检测各组PBMCs中GILZ mRNA的表达;ELISA检测核因子κB受体激活剂配体(RANKL)及骨保护素(OPG)的表达。结果:在10-7 mol/L DEX刺激下,DEX刺激组MM患者PBMCs的GILZmRNA表达水平高于空白对照组(P<0.05);DEX刺激组OPG的表达量高于对照组(P<0.05),而RANKL的表达量诱导组与空白对照组相比差异无统计学意义(P>0.05)。结论:糖皮质激素可有效上调GILZ基因表达;GILZ的高表达可以调节MM患者RANKL及OPG的平衡,具有改善溶骨性损害的作用。 Objective:To investigate the function of glucocorticoid induced leucine zipper(GILZ) in multiple myeloma.Method:Peripheral blood mononuclear cells(PBMCs) were collected from multiple myeloma patients by Ficoll and divided into two groups: dexmamethasone(DEX) treated group and control group.Forty eight hours after treatment,total RNA were extracted in both groups.Reverse transcription polymerase chain reaction(RT-PCR) was used to analyze the expression of GILZ mRNA in PBMCs from both groups.Enzyme linked immunosorbent assay(ELISA) was used to analyze the expression of RANKL and OPG protein.Result:Both OPG and GILZ were increased in DEX treated group compared with control group(P0.05).There is no difference of the expression of RANKL between two groups(P0.05).Conclusion:DEX is an effective stimulator of GILZ production.Overexpression of GILZ can regulate the balance between OPG and RANKL,suggesting that GILZ may have therapeutic value for bone diseases,such as multiple myeloma.
