摘要
目的阐明血吸虫病肝纤维化过程中,肝脏前胶原基因表达的动态变化规律。方法应用Northern核酸分子杂交技术,检测血吸虫感染小鼠在肝纤维化过程中肝脏前胶原mRNA表达量的变化。结果小鼠感染血吸虫后6周时,肝内α1(Ⅰ)和α1(Ⅲ)前胶原mRNA的表达量即有显著增加,8周时,α1(Ⅰ)前胶原mRNA的表达量已达高峰,α1(Ⅲ)和α1(Ⅵ)前胶原mRNA的表达高峰在10周时出现。12~16周时,三者的表达量略有下降,20周时均又回升。在整个肝纤维化过程中,α1(Ⅰ)前胶原mRNA呈优势表达,α1(Ⅲ)前胶原mRNA次之,α1(Ⅵ)前胶原mRNA的表达量较前二者稍低,但其表达水平的增幅最大。结论肝脏前胶原mRNA表达水平的检测能敏感地反映肝纤维化的趋向,α1(Ⅵ)前胶原mRNA的表达在肝纤维化形成中可能起重要作用。
Objective To
explore the dynamics of hepatic procollagen gene expression in the course of schistosomiasis
with liver fibrosis. Methods Northern blot hybridization analytical technique was used to
observe the dynamic changes of the content of liver procollagen mRNA in the course of liver
fibrosis in mice infected with Schistosoma japonicum. Results The expression of 1() and 1()
procollagen mRNA in the liver of mice was enhanced obviously at the 6th week after infection,
and the content of 1() mRNA reached to the peak at the 8th week. The content of 1() and 1()
procollagen mRNA both reached to their peak at the 10th week. The mRNA levels of all three
procollagens were decreased gradually at the 1216th week but rose again at the 20th week. The
expression of 1() procollagen mRNA was the most predominant one among the three in the
course of liver fibrosis. The content of 1() procollagen mRNA ranks next to 1() and that of 1()
procollagen mRNA was a little less than the other two, but its increment was much higher than
the two others. Conclusion Detection of the content of hepatic procollagen mRNA could
sensitively reflect the trend of liver fibrosis, and the expression of 1() procollagen mRNA maybe
play an important role in the pathogenesis of liver fibrosis.
