摘要
目的探讨肾络通药物血清对大鼠系膜细胞外基质不同成分的抑制作用及对转化生长因子β1(TGFβ1)表达的调节作用。方法以体外培养大鼠肾小球系膜细胞及血清药理学方法为基础,采用酶联免疫吸附试验检测细胞外基质中纤维连接蛋白(FN)、Ⅳ型胶原(ColⅣ)的含量;采用逆转录多聚酶链反应(RTPCR)检测TGFβ1mRNA的表达。结果细胞外基质FN,ColⅣ的含量与TGFβ1mRNA的表达呈正相关关系,肾络通对二者均有抑制作用。结论肾络通能够减少细胞外基质的积聚,抑制TGF-β1的分泌与表达,从而进一步明确了其防治多种肾小球疾病,延缓肾小球硬化的作用机制。
Objective: To investigate the effect of Shenluotong on the expression of transforming growth factor-β_1(TGF-β_1) and extracellular matrix(ECM) in Ang II-induced MCs. Method: Fibronectin(FN) and collagen type Ⅳ (Col Ⅳ) of extracellular matrix were detected by enzyme-linked immunosorbent assay; the expression of TGF-β_1 mRNA were measured by semi-quantitative reverse transcription-poly merase chain reaction. Result: A positive correlation btween TGF-β_1 and ECM were found in the present study. FN, ColⅣ and TGF-β_1 mRNA were inhibited by Shenluotong significantly. Conclusion: Shenluotong can decrease the accumulation of ECM and inhibit the expression of TGF-β_1,suggesting further that shenluotong can be used to prevent and treat various glomerular diseases and delay glomerular sclerosis.
