玫瑰黄链霉菌Men-myco-93-63固体发酵培养基的优化 被引量:5

摘要 研究玫瑰黄链霉菌Men-myco-93-63固体发酵培养基优化条件。采用单因素试验筛选的适宜得到固体基质为硅藻土,m(硅藻土)∶V(种子液)=2∶1,对菌落形成单位数有影响的因素为葡萄糖、蛋白胨、复合维生素和复合微量元素。在此基础上,通过响应面法对4个因素的组合进行优化,优化后这4个因素的质量分数最佳组合为葡萄糖2.2%、蛋白胨0.7%、复合维生素1.0‰、复合微量元素1×10-5。在此条件下,菌落形成单位数有了很大提高,达到1.7×108cfu/g。 The solid-state fermentation media of S.roseoflavus Men-myco-93-63 were studied in this research.Solid-state fermentation medium component of S.roseoflavus Men-myco-93-63,inoculation quantity were screened with mono-factor test.The result showed that suitable medium of solid-state fermentation was kieselguhr,and m(kieselguhr)∶V(seed culture fluid)= 2∶1,four factors impacted on the cfu(clonal formation unit) value of S.roseoflavus Men-myco-93-63,namely,glucose,peptone,compound vitamin and compound trace elements.Then,the optimal quality score of four factors were determined by response surface methodology,i.e.2.2%glucose,0.7%peptone,1.0‰ compound vitamin,1×10-5compound trace elements.The actual cfu value reached 1.7×108 cfu/g by using optimal medium.
