钠/碘转运蛋白和促甲状腺激素受体在甲状腺癌中的表达及相关性 被引量:3

Expression and correlation of sodium iodide symporter and thyroid stimulating hormone receptor in thyroid carcinoma

摘要 目的研究钠/碘转运蛋白(NIS)和促甲状腺激素受体(TSHR)在甲状腺癌中的表达特征,探讨二者表达的相关性。方法收集正常甲状腺组织18例、结节性甲状腺肿标本27例和甲状腺癌标本23例,通过荧光定量分析和免疫组化法研究NIS和TSHR的表达。结果甲状腺癌NIS和TSHR mRNA的表达显著低于正常甲状腺组织和结节性甲状腺肿(P<0.005)。在正常甲状腺组织、结节性甲状腺肿中,NIS和TSHR的表达呈显著正相关(r分别为0.667和0.551,P<0.005),而在甲状腺癌中无相关性。免疫组化结果表明在甲状腺癌中NIS阳性产物主要定位于细胞质,并显著高于正常甲状腺组织和结节性甲状腺肿(P<0.05);TSHR蛋白表达阳性产物主要定位于细胞质和细胞膜,显著高于正常甲状腺组织(P<0.001),但与结节性甲状腺肿无显著性差异。同正常甲状腺组织相比,甲状腺癌NIS mRNA表达下降的同时伴有细胞质NIS蛋白表达升高的比例显著高于结节性甲状腺肿(x^2=4.428,P=0.035)。结论甲状腺癌NIS mRNA及TSHR mRNA的表达下降,且二者表达无相关性。 Objective To investigate the expression of sodium/iodide symporter (NIS) and thyroid stimulating hormone receptor (TSHR) as well as their correlation in thyroid carcinoma. Methods mRNA levels, quantified by real-time flurescent quanti-tive polymerase chain reaction, and protein evaluated by immunohistochemistry, were examined in surgical specimens of 18 normal thyroid tissue, 27 nodular goiter and 23 thyroid carcinoma samples. Results The NIS and TSHR mRNA levels of thyroid carcinoma was significantly lower than in normal thyroid tissue and nodular goiter (P 〈0.005). There was a positive correlation between NIS and TSHR in nodular goiter and noral thyroid samples (r =0.667 and 0.551, respectively, P 〈0.005), there was no such correlation in thyroid carcinomas. The immunohistochemistry results of thyroid cancer showed that the NIS protein mainly located in cytoplasm, and was markedly higher than normal thyroid tissue and nodular goiter (P 〈0.05). TSHR protein mainly located in cytoplasm and cytomem- brane in thyroid carcinomas, and was significantly higher when compared with normal thyroid tissues (P 〈0.001), but had no difference with nodular goiter. Through combination analysis of NIS mRNA concentrations and NIS immunostaining scores, among the thyroid carcinomas, reduction of NIS mRNA expression but with increment of intracellular protein was markedly higher than in nodular goiter (χ2 =4.428, P =0.035). Conclusion In thyroid carcinoma, NIS and TSHR mRNA levels were low, also there was no significant correlation.
