摘要
目的:探讨消斑固斑片对兔颈动脉粥样硬化易损斑块崩解的机制。方法:取28-42日龄雄性SD大鼠,进行血管平滑肌细胞的原代培养,将培养细胞分为5组:空白对照组、模型组、消斑固斑片低剂量组、消斑固斑片中剂量组、消斑固斑片高剂量组。采用流式细胞术观察血管平滑肌细胞细胞间粘附分子-1蛋白表达(ICAM-1)。另高脂饲料加液氮法复制兔颈动脉粥样硬化模型,并随机分为对照组(10只),治疗组(10只),治疗组给予消斑固斑片(1.2 g/kg.d),连续灌胃15天,每天2次,两组均予高脂饮食,服药结束时。于处死动物前24 h、48 h给予中国斑点蛙蛇毒及组胺进行药物触发,以促使斑块发生实验性破裂。采用光镜观察斑块组织病理学和超微结构变化;采用酶法检测总胆固醇(TC)、三酰甘油(TG);磷钨酸-镁沉淀法检测高密度脂蛋白胆固醇(HDL-c)、低密度脂蛋白胆固醇(LDL-c)。结果:消斑固斑片有降低实验兔血中TC、TG、LDL-C,升高HDL-C的作用。同时可降低肿瘤坏死因子-α(TNF-α)诱导的ICAM-1水平及蛋白表达,且呈剂量依赖性。结论:消斑固斑片可通过降脂消斑稳定和修复易损斑块,延缓易损斑块的破裂作用。
Objective:To investigate collapse mechanism of the Xiaoban Guban Tablet on the vulnerable plaque of rabbit carotid artery scleratheroma.Methods:4 to 6 week-old male SD rats were given primary culture of the vascular smooth muscle cells and the cultured cells were divided into five groups:blank control group,model group,Xiaoban Guban Tablet of low-dose group,Xiaoban Guban Tablet of medium-dose group,Xiaoban Guban Tablet of high-dose group.To observe the expression of intercellular adhesion molecule-1 of smooth muscle cells through flow cytometry.In addition,to replicate carotid atherosclerosis models of rabbits with the method of high-fat diet with liquid nitrogen,which were randomly divided into the control group(10) and treatment group(10).The treatment group received Xiaoban Guban Tablet(1.2 g/kg·d),with continuous intragastric administration of 15 days,twice daily.Both groups are therefore correspondingly given high fat diet until the end of medication.24 h and 48 h before the animals were sacrificed,to give the Chinese spot frog venom and histamine for drug trigger to promote experimental rupture of the plaque.To observe plaque histopathology and ultrastructural changes by light microscopy;to test TC and TG by enzymatic detection,TG;to detect HDL-c,LDL-c through phosphotungstic acid and magnesium precipitation.Results:Xiaoban Guban Tablets have the effects of reducing TC,TG,LDL of the rabbits and increasing HDL in the serum.At the same time,it can lower the ICAM-1mRNA levels and protein expression induced by TNF-a induced in a dose dependent manner.Conclusion:Xiaoban Guban Tablet can stabilize and repair vulnerable plaque by lowering lipid and resolving plaques,thus delaying the rupture of vulnerable plaque.
