摘要
目的建立高脂血症大鼠模型,比较各时间点大鼠的血脂水平,为更合理的使用高脂血症动物模型提供依据。方法将96只SD大鼠随机分为普通饲料喂养组及高脂饲料喂养组(高脂饲料喂养组根据处死时间分为1d、2d、3d、4d、5d、6d、7d、14d、21d、28d、35d共11个亚组),每组8只大鼠。分别于相应时间点,对各组大鼠采血检测血脂水平,HE染色法观察肝脏的形态学变化。结果正常对照组随着时间增长,体重增长最快。各模型组大鼠的体重,在2d组与1d组,6d组与5d组,21d组与14d组相比较有统计学意义(P<0.05),其余各组相对于前一组均没有统计学意义(P>0.05)。模型各组大鼠TC、LDL-C与正常对照组比较均显著性升高(P<0.01),而TG、HDL-C则变化不明显。从造模1d起,大鼠血脂TC、LDL-C就开始升高,与正常对照组比较有显著性意义(P<0.01),并且随着时间推移逐步上升,从第4d到第21d保持相对稳定,第28d显著上升,至第35d开始下降。结论高脂饲料喂养法能在短时间内建立高脂血症大鼠模型,尤其对于高胆固醇型高脂血症及低密度脂蛋白型高脂血症效果明显。
Objective To explore the theoretical basis for a more reasonable use of hyperlipidemia animal model.Methods 96 SD rats were randomly divided into ordinary diet group and high-fat diet group(then divided into 11 subgroups according to the time when they were sacrificed: 1d,2d,3d,4d,5d,6d,7d,14d,21d,28d,35d).For each group,there were 8 rats.Blood lipids level were determined after the blood sampling.Hematoxylin-eosin staining was applied for observation on morphology of liver.Results The normal control group was observed a fastest growth in rat's weight,with statistical differences were significant between subgroups 2d and 1d,6d and 5d,21d and 14d(P0.05),whereas no obvious differences in the rest groups(P0.05).Total cholesterol(TC),low density lipoprotein-cholesterol(LDL-C) were elevated in the model groups in comparison with that in the normal group(P0.01),however with unobvious changes in triglyceride(TG) and high density lipoprotein-cholesterol(HDL-C).From the 1st day of modeling,the TC and LDL-C were elevating with time,with significant differences when compared with the normal control group(P0.01).From the 4th day to the 21st day there was no further elevation nor reduction in TC and LDL-C until the 28th day when another obvious elevation was observed which lasted to the 35th day when the indexes began to down-regulate.Conclusions The hyperlipidemia can be induced in rats with short time feeding with high fat diet,especially efficient for inducing hyperlipidemia of high cholesterol and low density lipoprotein.
