电导率法筛选微乳处方及对其相行为的研究 被引量:18

摘要 目的探讨电导行为结合伪三元相图制备微乳,并对其相行为进行研究。方法通过电导率-含水量曲线精确绘制聚山梨酯80-labrasol/Gemseal40-油酸/无水乙醇/水体系不同表面活性剂与助表面活性剂的比例(Km)下一系列伪三元相图,筛选微乳处方,采用电导率、稳定性、染色法、粒径等对微乳基本性能进行评价,研究不同Km对微乳体系的相行为及结构类型的影响。结果随着含水量的增加,电导率表现为渐进的连续变化,Km为2:1时,含水量(20%<Φ<66.3%)电导率直线上升,含水量(66.3%<Φ<74.5%)电导率趋势变缓,含水量(Φ>74.5%)时电导率下降,且此时伪三元相图中微乳区最大,制备丹皮酚微乳和雷公藤多苷微乳在高速离心无分层,也无絮凝或药物析出,电导率、染色法、粒径等性能均符合微乳特征。结论电导率法能用于微乳结构的判定并反映微乳相行为的变化,精确绘制伪三元相图筛选微乳处方。 OBJECTIVE To study the preparing microemulsion by conductance and pseudo-ternary phase diagram,and to research its phase behavior.METHODS According to conductivity-moisture content graph,a series of pseudo-ternary phase diagram about the system of tween80-labrasol / Gemseal40-oleic acid / anhydrous ethanol / water under the different proportion of surfactant and cosurfactant(Km) accurately,screening microemulsion prescription,evaluating basic performance of microemulsion with electric conductivity,stability,dyeing method,droplet size,studying the phase behavior and structure type of microemulsion system under different Km.RESULTS Along with the increase of water content,the conductivity showed gradual continuous change.When the value of Km was 2:∶1,the conductivity increased sharply with the water content range between 20% to 66.3%,the conductivity increased gently when the water content varied from 66.3% to 74.5%,the conductivity came up to the platform period and then declined while the water content was above 74.5%.Besides,the microemulsion area in pseudo-ternary phase diagram came to the largest at this ratio.Paeonolum-loaded and Tripterygium Glycosides-loaded microemusions were prepared.The microemulsion was stable under high speed centrifuge,electric conductivity,dyeing method,droplet size which was according with the characterization of microemulsion..CONCLUSION Conductivity can be used to determine the structure of microemulsions,and reflect the changes of phase behavior,it can screen microemulsion prescription by pseudo-ternary phase diagram.
