迷走神经结状神经节与心肌细胞联合培养的扫描电镜观察 被引量:1

SCANNING ELECTRON MICROSCOPICAL OBSERVATION OF CO-CULTURE OF NODOSE GANGLIA AND CARDIAC MYOCYTES

摘要 用新生大鼠的迷走神经结状神经节与心肌细胞进行联合培养,用扫描电镜观察了神经纤维或神经终末与心肌细胞之间的关系。在低倍下可见结状神经节组织块长出的神经纤维呈放射状向四周延伸,到达分散的心肌细胞表面;在扫描电镜高倍下所观察到的神经纤维或神经终末的形态不同,神经纤维粗细不一,在单个心肌细胞表面相互交织成网状。有的神经纤维在行程中粗细基本均匀一致.而有的神经纤维则在行程中出现膨体样结构。终止于心肌细胞表面的神经终末的形态也不相同,有的末端膨大终止于心肌细胞表面,有的末端膨大并呈扇形展开后紧贴于心肌细胞表面,这种膨大的神经末梢与心肌细胞之间的关系似比在体时更为密切。 To determine the relationship between nerve terminals from nodose ganglion and cardiac muscle cells, the samples ofco-culture of nodose ganglia and cardiac myocytes were observed with scanning electron microscope. Under asceptic conditions,hearts were removed from 6 newborn Sprague-Dawley rats (less than 24 hours after birth ). From the newborn rats, bilateral nodose ganglia were removed separately but simultaneously. Dispersion culture method was used for cardiac muscle cells. Organized culture method was used for nodose ganglia. The cardiac muscle cell suspension, at a density of 106 cells/ml, was plantedin petri dishes, each containing a sterile glass coverslip (24 mm in diameter). The coverslips, prior to cardiac muscle cell planting, contained 3 or 4 nodose ganglia explants. After 96 hours of incubation at 37℃ in a 5 % CO2 incubator, the neuromuscularco-cultures were observed with a Hitachi S-520 scanning electron microscope. At low magnifications, nodose ganglion explantssent radial projections to the surface of cardiac muscle cells. At high magnifications, the form of the nerve fibers or terminals observed were variable. The nerve fibers were different in caliber and formed a network on the surface of some cardiac muscle cells.Some nerve fibers extended directly from the immediate proximal neuronal processes, some were widened into varicosities. Someendings appeared to be fan--shaped and tightly terminated on cardiac muscle cell surface. The result suggested that in vitro the reintionship between sensory nerve endings and cardiac muscle cells was closer than that in vivo. The establishment of this modelmight open a new area to study the relationship between sensory nerves and cardiac muscle cells.
