摘要
目的:研究大黄素与DNA相互作用。方法:通过共振光散射光谱和荧光光谱,发现大黄素与DNA相互作用产生强烈增强的共振光散射和荧光信号。结果:DNA与大黄素作用在556nm处产生较强共振散射峰,在540nm有很强的荧光峰,且荧光和散射强度均呈良好的线性关系。结论:通过Scatchard方程和双倒数曲线得出大黄素与DNA的结合常数是KA=0.18×105L·mol-1,说明它能嵌入到DNA碱基对之间形成复合物,也是一种DNA嵌入剂。
Objective:The interaction of emodin and DNA was investigated by using RLS spectra and fluorescence spectra.Method:The interaction of emodin and DNA results in strongly enhanced resonance light scattering spectra characterized by peak.Result:It was found that the enhanced RLS intensity at 556 nm was proportional to the concent ration of emodin and the enhanced fluorescence intensity at 540 nm was proportional to the concent ration of DNA,and there are certain fluorescence spectra and RLS linear relationship.Conclusion:Figure the combination constant of emodin and DNA KA=0.18×105L·mol-1 by Scatchard plots.The result can show that it can be plugged into DNA base and form compounds,also is a kind of DNA intercalator.
