摘要
【目的】研究秦川牛生长激素基因第2外显子的多态性,及其与部分体尺性状指标的相关性。【方法】以502头2周岁左右秦川牛为研究对象,利用PCR-RFLP技术检测生长激素基因上存在的单核苷酸多态性(SNPs),并分析其与秦川牛体尺性状的关联性。【结果】在GH基因第5外显子中发现了2处SNPs,分别为错义突变和同义突变。位点1经AluⅠ酶切后产生AA、AB和BB 3种基因型,A、B等位基因频率分别为0.825 7和0.174 3;卡方检测显示,该位点在所检测秦川牛群体中处于Hardy-Weinbery不平衡状态;关联分析表明,该位点不同基因型个体之间在体高、胸围、体斜长和尻长上差异显著。位点2经AflⅡ酶切后产生CC和CD 2种基因型,C和D等位基因的频率分别为0.902 4和0.097 6;卡方检测显示,该位点在所检测秦川牛群体中处于Hardy-Weinbery不平衡状态;关联分析表明,该位点不同基因型个体间在腰角宽、坐骨端宽和尻长上差异显著。第5外显子2个位点合并基因型分析结果表明,双杂合基因型个体各体尺性状均显著高于其他基因型组合,并且多标记位点的组合效应高于单标记位点。【结论】组合ABCD各体尺性状数值均高于AB和CD基因型,组合ABCD可能是影响牛体尺性状的最佳基因型组合。
【Objective】 The research was to study the relationship of the polymorphism in exon 5 of GH gene on several body measurement traits in Qinchuan beef cattle.【Method】 502 Qinchuan cattles(Bos tarurs) aged 2 years were selected to detect polymorphism in exon 5 by using PCR-RFLP,and to study its associations with Qinchuan cattle body measurement traits.【Result】 The result showed that two mutations were found in exon 5,which were missense mutation and synonymous mutation.There were 3 genotypes named AA,AB and BB respectively after digestion by AluⅠ.Allele frequency of A and B were 0.825 7 and 0.174 3 respectively.χ2 test showed that the locus was not in Hardy-Weinberg equlibrium in the Qinchuan cattle population detected.Correlation analysis showed that individuals with different genotypes had significant differences in wither height,heart girth,body length and rump length.There were 2 genotypes named CC and CD respectively after digestion by AflⅡ.Allele frequency of C and D was 0.902 4 and 0.097 6 respectively.χ2 test showed that the locus wasn't in Hardy-Weinberg equlibrium in the Qinchuan cattle population detected.Correlation analysis showed that individuals with different genotypes had significant differences in hip width,ischium width and rump length.The result of analysis of two locuses combined genotypes in exon 5 showed that body measurement traits of combined genotype ABCD in exon 5 were higher than those of the other kinds of combinations,and the effects of multi-locus combinations of marker were higher than those of single-marker locus.【Conclusion】 The efficiency of combined genotype was better than that of single-marker locus.
