摘要
采用PCR-SSCP及DNA测序技术,以106头信阳水牛为材料,采集血样,提取基因组DNA,以GenBank公布的牛GH基因序列(NC_007317)为参照序列,设计PCR引物,对GH基因的第2、3外显子及部分内含子进行了遗传变异检测,并将其多态性与部分生长性状进行了相关分析。结果表明:每个位点的PCR产物经SSCP分析均出现不同的带型,第2外显子存在1处SNP(G413A),第2内含子存在4处SNPs(G495C,A523C,A541G,G699A),信阳水牛GH基因的GH-P3位点的多态性与部分生长性状没有显著相关性。
Blood samples from 106 xinyang buffalo were extracted for genomic DNA and primers for Gtt gene were designed to detect the polymorphisms in the second and third exon and part of introns of GH gene by using PCR - SSCP and DNA se- quencing technology. Then the correlations between the polymorphisms and growth traits in Xinyang buffalos were analyzed. The results showed that in the tested buffalos each locus had shifted SSCP band and there was one SNP(G413A) in the sec- ond exon and four SNPs (G495 C, A523C, A541 G, G699A)in the second intron. The correlation analysis results showed that the genetic polymorphisms in GH - P3 locus of GH gene had no significant correlation with growth traits in xinyang buffalo.
