摘要
目的:探讨羟基红花黄色素A(Hydroxysafflor yellow A,HSYA)对人主动脉内皮细胞(Human aortic endothelial cells,HAEC)血小板反应蛋白1(Trombospondin-1,TSP-1)表达的影响,以进一步研究HSYA抑制内皮细胞增殖及抗肿瘤的作用机制。方法:采用四甲基偶氮唑蓝(MTT)法观察HSYA对HAEC增殖作用的影响。Real Time PCR法及免疫细胞化学法分别检测HSYA作用后,HAEC内TSP-1的mRNA及蛋白表达;ELISA法检测上清液中TSP-1含量。结果:HSYA对HAEC增殖作用的影响表现出浓度依赖性,较高浓度(>0.0657g/L)对HAEC的增殖有抑制作用,较低浓度(<0.0657g/L)表现出促进作用。HSYA对TSP-1mRNA表达有促进作用,且这种作用随浓度增加而增强;TSP-1的蛋白表达与释放情况与RT-PCR结果相似。结论:HSYA能促进HAEC表达TSP-1;HSYA对内皮细胞的增殖表现出促进和抑制的双向作用,可能是TSP-1和血管内皮生长因子(Vascular endothelial growth factor,VEGF)等多种因素共同作用的结果。而HSYA的抗肿瘤作用可能是通过较高浓度HSYA提高TSP-1的表达抑制内皮细胞增殖进而抑制血管形成来实现的。
Objective:To investigate the influence of Hydroxysafflor yellowA(HSYA)on the expression ofthrombospondin-(lTSP-1)in human aortic endothelial cells for a further study of the mechanism of hydroxysafflor yellow A inhibiting endothelial cells'proliferation and preventing the growth of tumor.Methods:We use the method of MTT to observe the proliferation of HAEC with the influence of HSYA.Real Time PCR and immunocytochemistry were used to detect the level of mRNA and protein of TSP-1 after HSYA's effect.TSP-1 in the culture medium is assayed in the way of ELISA.Results:HSYA's effect on proliferation of HAEC showed a concentration-depen dence,for higher concentration(0.065 7 g/L)causing inhibition of the proliferation of HAEC while a lower concentration(0.065 7g/L)arosing catalyst performance.HSYA promotes the expression of mRNA of TSP-1,and the effect is enhanced as the concentration increases.;The expression of TSP-1 is similar to the result of our RT-PCR.Conclusion:HSYA promotes the expression of TSP-1.The HSYA shows a two-way effect on the proliferation of endothelial cells,and regulating the expression of TSP-1and VEGF may be the main way to achieve it.The anti-tumor effect of HSYA may be through a way of promoting the expression of TSP-1 to inhibit the proliferation of endothelial cells and prevent angiogenesis.
