摘要
在单因素实验和多因数-二水平实验的基础上,采用Box-Benhnken实验设计,选取水溶性淀粉、酵母浸膏和培养时间为自变量,以荧光物质的总荧光强度作为因变量,通过响应面法优化了罗汉果内生真菌SG-59产荧光物质的培养条件。结果表明,此菌株产荧光物质的最佳培养条件为:水溶性淀粉36.5 g/L,酵母浸膏7.0 g/L,500 mL培养瓶装液量为170 mL,28℃静置培养27 d。这将为大规模发酵和进一步研究这些荧光物质打下基础。
Based on the single factor experiments and multi-factors-two-level experiments, the water- solubility starch, yeast extract, and cultivated time were selected as independent variables and the total fluorescence intensity was as the dependent variable using Box-Benhnken experimental design Finally the culture medium of a strain endophytic fungus SG-59 from Siraitia grosvenori producing fluorescent substance was optimized by response surface methodology. Results showed that the best culture conditions were as follow : water-solubility starch 36.5 g/L, yeast extracts 7. 0 g/L, 500 mL bottle culture filled with 170 mL, static culture 27 days under the temperature of 28 ℃. This will lay foundations for the further study of those fluorescent after large-scale fermentation.
