冬虫夏草及人工虫草丝菌提取物对四氯化碳致急性化学性肝损伤小鼠的保护作用 被引量:2

Protective effects of cordyceps sinensis and mycelium of cultured sinensis on acute liver injury and expression of CYP450 induced by Carbon Telrachloride

摘要 目的研究冬虫夏草及人工虫草丝菌对四氯化碳(CCl。)致急性化学性肝损伤小鼠细胞色素P450(CYP450)活性的影响,探讨其对肝损伤的保护作用机制。方法昆明种雄性小鼠CCl4(0.1%,20ml/b)一次性腹腔注射建立急性化学性肝损伤模型,冬虫夏草及人工虫草丝菌(100mg/m1)每6h灌胃一次,共3次,随后在3个不同时间段(6h、12h、24h)处死小鼠,检测血清ALT、GSH及肝微粒体CYP450含量,Westernbolt检测CYP450蛋白表达。结果冬虫夏草及人工虫草丝菌在各个不同时间段明显降低ALT水平,以24h降低更为明显,且GSH、CYP450活性提高。结论冬虫夏草及人工虫草丝菌对化学性肝损伤治疗作用,可能与其提高CYP450等肝药酶活性,增加其抗氧化作用有关。 Objective To investigate the effect of Cordyeeps Sinensis and Myeelium of Cultured Sinensis on acute injury in this test and expression of CYP450. Methods Mice received a single dose of 0. 1% CC14 i. p, after CC14 intoxication,mice received Cordyeeps Sinensis and Myeelium of Cultured Sinensis (100mg/ml) 3 times per six hour,then the mice sacrificed at 6h, 12h,24h ,the levels of aminotransferase (ALT) and glutathioue (GSH) in ser- um, CYP45 content in hepatle mierosomal aetlvity in liver homogenate were measured. Results Mice given Cordyeeps Sinensis and Myeelium of Cultured Sinensis supplement had less elevation of serum ALT concentration, Cordyeeps Sinensis and Myeelium of Cultured Sinensis administration restored the reduction of hepatic mierosomal P450 protein content as well as inducing an increase in hepatic GSH concentration. Additionally, Cordyeeps Sinensis and Myeelium of Cultured Sinensis treatment inhibited the elevation of bepatie CYP450 protein after CC14 intoxication. Conclusion The proteet effeet of Cordyeeps Sinensis and Myeelium of Cultured Sinensis may be due to its ahilitity to inerease CYP450 eontent which deereased in CCI4-indueed liver in combination with increasing tile activities of antioxidant en- zyme.
