食管鳞状细胞癌组织中乏氧诱导因子-1α蛋白和mRNA的表达 被引量:2

Expression of HIF-1α protein and mRNA in esophageal squamous cell carcinoma tissue

摘要 目的:探讨食管鳞癌组织中乏氧诱导因子-1α(HIF-1α)的表达及其临床意义。方法:分别采用免疫组化及RT-PCR技术检测46例食管鳞癌组织和46例正常食管黏膜组织中HIF-1α蛋白及mRNA的表达情况,并分析2者的表达与食管鳞癌患者临床病理特征的关系。结果:食管鳞癌组织中HIF-1α蛋白阳性染色主要位于癌细胞胞质/核内,阳性率为41.3%(19/46),而正常食管黏膜组织中均呈阴性表达。食管鳞癌组织中HIF-1αmRNA的表达水平(1.17±0.25)高于正常食管黏膜组织(0.35±0.12)(t=13.942,P<0.001)。46例食管鳞癌组织中HIF-1αmRNA高表达(高于癌组织中的平均水平)者为24例(52.2%)。食管鳞癌组织中HIF-1αmRNA表达水平与HIF-1α蛋白表达阳性率呈正关联(rP=0.474,P<0.01)。食管鳞癌组织中HIF-1α蛋白的表达与肿瘤的浸润深度(χ2=6.624,P=0.010)和淋巴结转移有关(χ2=7.404,P=0.007),HIF-1αmRNA表达水平与肿瘤的组织学分级(χ2=0.321,P=0.571)、淋巴结转移(χ2=0.708,P=0.400)、浸润深度(χ2=2.333,P=0.127)及临床分期(χ2=0.067,P=0.800)均无关。结论:食管鳞癌组织中HIF-1α在蛋白水平的表达对临床更具有指导意义。 Aim : To investigate the expression of HIF-1α and its significance in esophageal squamous cell carcinoma (ESCC) tissue. Methods :The expression of HIF-1α on mRNA and protein levels was investigated respectively by reverse transcriptase polymerase chain reaction(RT-PCR) and immunohistochemistry in 46 cases of ESCC tissue and 46 cases of normal mucosal tissue of esophagus. The relationship between the expression level of HIF-1α and clinicopathological features was investigated in ESCC. Results: HIF-1α immunoreactivity was recognized in both cytoplasm and nuclei. The positive expression rate of HIF-1αwas 41.3% (19/46) in ESCC tissue. And it was negative expressed in normal tissue. The mRNA expression the level in ESCC tissue( 1. 17 ± O. 25 )was significantly higher than that in normal tissue(0.35 ± 0. 12) ( t = 13. 942 ,P 〈0. 001 ). High level HIF--1α mRNA ( more than 1.17) was detected in 24 out of 46 tumor cases. The high expression rate was 52.2%. There was positive correlation between the level of HIF-1α mRNA expression and the positive rate of HIF--1α protein expression (re = 0. 474 ,P 〈0.01 ). HIF-1α protein expression in ESCC tissue was related with in- vasion depth(x2 = 6. 624 ,P = 0. 010) and lymph node metastasis(x2 = 7. 404, P = 0. 007 ). The mRNA expression level of HIF-1α was not associated with histological grades (X2 = 0. 321, P = 0. 571 ) , lymph node metastasis (X2 = 0. 708, P = 0. 400), infiltration depth (X2 = 2. 333, P = O. 127 ), or clinical stage (Xz = 0. 067, P = 0. 800 ). Conclusion : It indicates that HIF-1α protein can be used as a new indicator in ESCC.
