纤维素酶水解漂白硫酸盐蔗渣浆的研究

摘要 本课题以硫酸盐漂白蔗渣浆为原料采用C1184纤维素酶粉作为水解酶,探讨了甘蔗渣浆酶法水解的最佳工艺条件,研究了反应时间、酶用量、反应温度、反应体系的pH值,底物粒度等因素对纤维素酶水解糖化甘蔗渣的影响。结果表明,甘蔗渣浆酶法水解的最佳条件为:原料粒度为100目,在pH值为4.5的乙酸—乙酸钠缓冲液存在的体系中,与0.23 g活性为8418 IU/g的C1184纤维素酶在温度为55℃的条件下,在全温振荡器中反应44 h后,用紫外-可见分光光度计分析还原糖含量,在最优化条件下,蔗渣浆酶法水解的总转化率为75.45%。正交分析法表明,影响蔗渣浆纤维素酶解的最主要因素是pH值,其次是酶用量和温度,反应时间的影响最小。 Bleached Kraft bagasse pulp and C1184 cellulase were used to study the best technique conditions on the enzymatic hydrolyzing BKP.The effect of reaction time,dosage of reaction temperature,pH value of reaction system and bottom size on bagasse pulp hydrolyzed by cellulose was also studied.The results showed that the bagasse pulp with size of 100 head,the pH of system in acid-sodium acetate buffer was 4.5 and the dosage of C1184 with enzyme activity 8418 IU/g was 0.23 g,the reaction temperature was 55℃ and the reaction time was 44 hours.The content of reducing sugar was analyzed by Vltraviolet-Vis when the reaction ended.At the most optimum conditions,the total yield of bagasse pulp enzymatic hydrolysis was75.45%.By orthogonal analysis,the most important factor in the reaction was the pH value,followed by the enzyme dosage and temperature,the reaction time was the least.
