摘要
目的:研究中药夏枯草提取物(Prunella vulgaris Linn.extract,PVE)联合化疗药物对人淋巴瘤Raji细胞增殖的影响。方法:MTT法检测PVE分别联合紫杉醇(paclitaxel,TAX)、多柔比星(adriamycin,ADM)对Raji细胞的生长抑制率;FCM法检测PVE与TAX联合对Raji细胞周期分布及凋亡的影响;免疫组织化学法检测PVE对Raji细胞内survivin及caspase-3蛋白表达的影响。结果:PVE、TAX及ADM对淋巴瘤Raji细胞的生长均表现出明显的抑制作用,低剂量PVE在0.8~0.0008μg/mL浓度范围内可增强TAX对Raji细胞的抑制作用(P<0.01);在0.08~0.0008μg/mL浓度范围内与ADM联合使用时细胞生长抑制率较单用ADM时明显增高(P<0.01)。与TAX单用组比较,PVE与TAX联用组的S期细胞数下降,G2/M期细胞数明显升高。免疫组织化学检测结果显示,PVE(30μg/mL)可使Raji细胞中caspase-3蛋白表达增强,survivin蛋白表达减弱。结论:PVE可增强Raji细胞对TAX及ADM的敏感性。
Objective:To investigate the effects of extract from Prunella vulgaris Linn. (PVE) combined with chemotherapeutic agents on proliferation of lymphoma cells. Methods:The growth inhibition rate of Raji cells treated with PVE combined with paclitaxel (TAX) and adriamycin (ADM) were examined by MTT assay. The distribution of cell cycle and apoptosis of Raji cells were tested by flow cytometry. The expressions of apoptosis related proteins survivin and caspase-3 were detected by immunocytochemistry. Results:PVE,TAX,and ADM obviously inhibited the proliferation of Raji cells. PVE at 0.8-0.000 8 μg/mL enhanced the sensitivity of Raji cells to TAX,and the inhibitory effects of PVE at 0.08-0.000 8 μg/mL combined with ADM were significantly higher than that induced by ADM alone (P〈0.01). As compared with TAX group,PVE+TAX resulted in a reduction of the cell proportion in S phase but an increase in G2/M phase. Immunohistochemistry showed that PVE 30 μg/mL up-regulated the expression of caspase-3 and down-regulated the protein expression of survivin. Conclusion:PVE could enhance the sensitivity of Raji cells to TAX and ADM.
