摘要
目的:建立HPLC测定中药黄山药中伪原薯蓣皂苷含量的方法。方法:采用反相高效液相色谱法,Merck Puro-spher STARRP-18e色谱柱(4.6mm×250mm,5μm);流动相为乙腈-水梯度洗脱,流速1mL·min-1,检测波长203nm,柱温40℃。结果:伪原薯蓣皂苷在此色谱条件下获得良好分离,含量测定的线性范围0.08~4.00μg(R2=0.9999),平均加样回收率为99.9%,RSD2.3%。结论:首次建立了高效液相色谱法测定黄山药药材中伪原薯蓣皂苷含量的方法,该方法准确、可靠,可用于黄山药药材及其制剂质量控制的方法。
Objective:To develop an HPLC method for the determination of pseudoprodioscin in Dioscorea panthaica and provide scientific basis for quality control of it and its preparation. Method:The analysis was carried out on a Merck Purospher STAR RP-18e column eluted with acetonitrile and water as mobile phases in gradient mode. The detection wavelength was 203 nm,and the flow rate was 1.0 mL·min-1. Result:The pseudoprodioscin in D. panthaica extract was well separated. The linear range is between 0.080 0-4.00 μg,r=0.999 9. The average recovery and RSD are 99.9% and 2.3%,respectively. Conclusion:This method for quantitation of pseudoprodioscin in D. panthaica is simple,accurate and reliable and can be used for the quality control of D. panthaica and its preparations.
