摘要
目的:探讨蟾酥抗链球菌感染的物质基础。方法:采用链球菌感染及链球菌外毒素刺激人外周血单个核细胞(PB-MC)增殖模型,对蟾酥、蟾酥化学提取部位及主要蟾蜍甾烯进行活性筛选。结果:蟾酥脂溶性部位显著抑制链球菌感染小鼠的死亡率,显著抑制链球菌体外增殖。蟾酥脂溶性部位及所含三种主要蟾蜍甾烯(脂蟾毒配基、华蟾毒精、蟾毒灵)均显著抑制链球菌毒素刺激人PBMC的增殖,而水溶部位无明显作用。结论:含有蟾蜍甾烯的脂溶性部位是蟾酥抗链球菌感染的主要活性部位。
Objective: Venenum bufonis,a famous traditional Chinese medicine for treatment of infectious inflammation,was evaluated for its anti-infectious activities.Methods :Streptococci infection model in mice and Streptococci proliferation in vitro were used to test anti-infectious activity.Results: It was observed that Venenum bufonis(16mg/kg) and its chloroform fraction(4mg/kg) significantly improved survival in mice injected by group A streptococcus(GAS).Venenum bufonis chloroform fraction further inhibited GAS proliferations in vitro with IC50=5.65μg/ml. Venenum bufonis (50 -500 ng/ml) and its chloroform fraction (10 - 100 ng/ml) potently inhibited proliferation of human peripheral blood mononuelear cell (PBMC) stimulated by streptococcal pyrogenic exotoxin in vitro. In addition, three bufadienolides in chloroform fraction (resibufogenin, cinobufagin, bufalin) all had remarkable inhibitive actions on PBMC proliferation. In contrast, Venenum bufohis water fraction had no these activities above mentioned. Conclusion : These results suggest that Venenum bnfonis has significantly anti- GAS-infectious activities. The chloroform fraction is a key fraction of Venenum bufonis to the overall anti-GAS-infectious activities effects.
