高效液相色谱电化学法测定人血清中的儿茶酚胺及5-羟色胺含量 被引量:8

Determination of catecholamines and 5-HT in human serum by HPLC-ECD

摘要 目的:建立高效液相色谱电化学法(HPLC-ECD)测定人血清中的儿茶酚胺(NE、E、DA)及5-羟色胺(5-HT)含量的方法。方法:运用高效液相色谱分离及电化学检测器检测,氧化铝固相萃取法测定NE、E、DA,5-磺基水杨酸蛋白沉淀法测定5-HT。结果:NE、E、DA在1-10 ng/ml,5-HT在10-100 ng/ml范围内线性良好(n=5,r均>0.998),样品加标回收率为88%-105%,方法最低检出限:NE、E、DA为0.1 ng/ml,5-HT为1 ng/ml。结论:高效液相色谱电化学法(HPLC-ECD)测定人血清中的NE、E、DA及5-HT含量,该方法灵敏度高,回收率和重现性良好,样本用量少,操作快速简便,结果准确可靠。 Objectives:To establish a method to detect catecholamines such as nore-pinephrine(NE),epinephrine(E) and dopamine(DA),and 5-hydroxy tryptamine(5-HT) in human serum by high-performance liquid chromatography-electrochemical detection(HPLC-ECD).Methods:HPLC and electrochemical detector were used to assay catecholamines and 5-HT in this research.Solid-phase extraction of alumina was applied for determination of NE,E and DA in human serum were determined by,while sulfosalicyclic acid was used as protein precipitator for determination of 5-HT in human serum.Results:Result of NE,E and DA showed a good linear relationship at the range of 110 ng/ml,and result of 5-HT indicated a good linear relationship at the range of 10100 ng/ml(n=5,r〉0.998).The average recovery ratio was 88%105%.The limit of detection for NE,E and DA was 0.1 ng/ml,while that of 5-HT was 1 ng/ml.Conclusion:HPLC-ECD is a stable,highly sensitive and reproduciable method to determine NE,E,DA and 5-HT in human serum.
