摘要
利用高效液相色谱法测定了吉林省500份大豆品种(系)中的低聚糖含量(干基)。使用正相氨基柱分离、63%乙腈溶液洗脱、示差折光检测器检测,以蔗糖、水苏糖和棉籽糖为标准品,加入乳糖为内标,计算大豆中低聚糖含量。蔗糖、水苏糖和棉籽糖的加标回收率分别为99.85%、100.34%和98.21%,线性相关系数为0.9987~0.9996。测得500份大豆品种(系)中的低聚糖总量范围7.73%~13.46%,超过12%的品种(系)有49个。蔗糖含量范围3.42%~8.27%,棉籽糖含量范围0.41%~1.37%,水苏糖含量范围3.23%~4.87%。
The contents of the oligosaccharides in 500 soybean varieties (lines) in Jilin province were determined by high performance liquid chromatography. The sucrose, raffinose, stachyose were used as standards, and the lactose was used as internal standard. The oligosaccharides were eluted, separated, and detected by 63 % acetonitrile solution, NH2 column and refractive index detector, respectively. The recoveries of standard spiking of 3 oligosaccharides( sucrose, raffinose and stachyose) were 99.85 %, 100.34% ,98.21%, the relative standard deviation( RSD, n = 3 ) were 1.86 % ,2.32 % ,3.43 % , respectively. There was a good linear relationship between concentration ( μg.mL^- 1 ) and peak area for these three oligosaccharides. The linear correlation coefficient ranged form 0.9987 to 0. 9996. The sucrose, raffinose , stachyose content ranged from 3.42 % to 8.27 % ,from 0.41% to 1.37 % ,and from 3.23 % to 4.87 % ,respectively,and the total soybean oligosaceharides content ranged from 7.73 % to 13.46 %. The total oligosaeeharides content of 49 varieties(lines) soybean exceeded 12 % and the sucrose content of 29 varieties (lines)soybean exceeded 7 %. Results indicated that varietal differences in oligosaccharides content of soybean varieties (lines)were highly significant. The oligosaccharides content of soybean varieties ( lines ) in Jilin province were commonly higher. The research results lay a foundation for the further research of soybean quality breeding and processing and utilization of soybean oligosaccharide.
