细胞周期和bcl-2、bax表达与脑胶质瘤病理分级的相关性研究 被引量:6

摘要 目的:探讨细胞周期和凋亡相关蛋白bcl-2和bax与脑胶质瘤病理分级的相关性,为脑胶质瘤严重程度的临床评价和机制研究提供参考依据。方法:取99例脑胶质瘤组织,分离获得单个胶质瘤细胞,采用FCM进行细胞周期和bcl-2及bax的测定,同时收集相应的病理分级资料,进行相关性分析和多元逐步回归分析。结果:病理分级与细胞周期(S期)及bcl-2、bcl-2/bax均呈正相关性,随着脑胶质瘤病理分级程度的升高,S期比例上升(t=2.48,P=0.018 4,R2=0.90),bcl-2、bcl-2/bax比值均上升(t=23.26,P<0.000 1,R2=0.93;t=27.42,P<0.000 1,R2=0.85)。结论:细胞周期S期分析和bcl-2、bcl-2/bax在脑胶质瘤严重程度的临床评价和机制研究中具有一定的参考价值。 Objective:To study the relationship between cell cycle, bcl-2 and bax expressions and the pathological grades of glioma in order to provide help for clinical evaluation and mechanism research of the severity of human glioma. Methods:This study collected 99 human glioma samples and isolated single glioma cells. Flow cytometry was used to detect their cell cycle and determine the expression levels of bcl-2 and bax. The pathological grading data were collected concurrently. The correlation and multiple stepwise re- gression analysis were analyzed by SAS software. Results:Pathologic grades had positive correlation with the percentage of glioma cells in S phase, the level of bcl-2, and the value of bcl-2/bax. As the increase in the pathologic grade, the percentage of glioma cells in S phase was also increased (t = 2.48, P=0.018 4, R2 = 0.90), the expression level of bcl-2 and the ratio of bcl-2/bax were elevated (t = 23.26, P 〈 0.000 1, R2 = 0.93 ;t = 27.42, P 〈 0. 000 1, R2 = 0.85 ). Conclusion: The percentage of glioma cells in S phase, the level of bcl-2, and the ratio of bcl-2/bax had some value for clinical evaluation and mechanism research of severity of glioma.
