摘要
目的:探讨姜黄素对类风湿关节炎患者滑膜细胞增殖与凋亡的影响。方法:用酶消化法分离类风湿关节炎患者膝关节滑膜细胞,四甲基偶氮唑蓝(MTT)法测定5,10,20μmol.L-1姜黄素作用48,96,144h后对滑膜细胞增殖的抑制率,流式细胞仪定量检测3种浓度姜黄素干预48h后细胞的凋亡率,半定量逆转录聚合酶链反应法检测细胞bcl-2、bax mRNA的表达。结果:5,10,20μmol.L-1的姜黄素分别干预48,96,144h后滑膜细胞的增殖均受到不同程度的抑制,抑制程度与药物浓度呈剂量依赖性。3种浓度的姜黄素干预48h后均可明显诱导滑膜细胞的凋亡,凋亡率均明显高于空白对照组(P<0.01)。3种浓度姜黄素均可提高bax/bcl-2mRNA的表达,10,20μmol.L-1姜黄素组明显高于空白组(P<0.01)。结论:姜黄素对体外培养的类风湿关节炎患者滑膜细胞的增殖具有一定的抑制作用,并可诱导其早期凋亡,可能与上调bax/bcl-2mRNA的表达有关。
OBJECTIVE To investigate the effect of curcumin on the proliferation and apoptosis of synoviocyte isolated from the patients with rheumatoid arthritis. METHODS The synoviocyte from the knee joint of patient with rheumatoid was isolated by enzyme digestion. The inhibition effect on the third passage cell proliferation was assayed by MTT method after Being interfered 48,96,144 h by 5, 10, 21） μ mol· L^-1 curcumin, respectively. The cell apoptosis rate were analyzed by flow cytometry （FCM） after being interfered 48 h. The expression of bcl-2 mRNA and bax mRNA was measured by semi-quantitative RT-PCR method. RESULTS After being interfered 48 h.96 h, 144 h,the cell proliferation was inhibited to different extent on the basis of the curcumin concentration, respectively. Compared with the control group, the level of apoptosis rate was significantly incresed in 5,10,21μmol· L^-1 curcumin group （P〈0.01）, respectively. The expression ratio of synoviocyte bax mRNA/bcl-2 mRNA was incresed to different extent by every curcumin concentration, as well,and were significantly higher in the 10,20 μmol· L^-1 curcumin groups than that of control group （P〈0. 01 ）. CONCLUSION Curcumin can inhibit proliferation and induce apoptosis of synoviocyte isolated from the knee joint of patient with rheumatoid in vitro. This effect may be related to up-expression of bax/bcl -2 mRNA by curcumin.
