摘要
目的:提高盾叶薯蓣试管苗的移栽成活率。方法:在组织培养的基础上,对盾叶薯蓣人工种子中人工种皮、人工胚乳制作、人工种子一次性成苗及移栽成活率等问题进行了初步研究。结果:人工种子快速繁殖的最适条件为:从盾叶薯蓣愈伤组织表面切取2~3mm小块茎,包埋在2.5%海藻酸钠中,滴入2%CaCl2中反应8~10min,人工胚乳MS/2+IAA0.4mg/L直接播种在MS/2培养基上,一次性成苗率可达100%左右,移栽成活率高达85%。结论:以组织培养为基础的盾叶薯蓣人工种子快速繁殖技术具有一定的优越性。
Objective: To improve the survival rate of the plantlets of D.zingiberensis. Methods: On the basis of tissue culture, the technique of artificial endosperm, artificial seeds of D.zingiberensis, the germination ratio of artificial seeds and the survival rate were investigated. Results: The condition of rapid multiplication of artificial seeds should be as following: cutting about 2-3 mm size embryo coming from callus, cultured embryo was encapsulated with 2.5% solidumalginate in IAA 0.4 mg/L solution and then dropped in 2% CaCl2 solution for 8-10 min, artificial seeds were germinated on the MS/2 medium. According to the above procedure, the germination ratio of artificial seeds could reach 100% or so and the survival rate could reach 85%. Conclusion: The technique of rapid multiplication of artificial seeds on the basis of tissue culture of D.zingiberensis has definite superiority.
