摘要 以杂种白杨‘717杨’(Populus tremula×Populusalba)和‘353杨’(Populustremula×Populus tremuloides)的叶片为材料,研究不同基因型、激素组合对叶片分化和不定芽生根的影响。结果表明:‘717杨’叶片最适分化培养基是MS+0.5mg/L6-BA+0.2mg/LNAA,‘353杨’叶片最适分化培养基是MS+0.5mg/L 6-BA+0.2mg/LNAA+0.01mg/LTDZ;最佳生根培养基为1/2MS+0.2mg/LIBA+0.2mg/LNAA,生根率达100%;叶片培养的最佳位置是自茎尖展叶的1~3片叶;20mg/L卡那霉素可以抑制‘717杨’和‘353杨’叶片的诱导分化,40mg/L卡那霉素可以抑制‘717杨’和‘353杨’不定芽的生根。 The regeneration of adventitious buds and the rooting of shoots from the leaves of hybrid 717 from Populus trernula × Populus alba and 353 from Populus trernula × Populus trernuloides with different genotypes and hormones combination were studied. The results showed that: the optimum medium for ad- ventitious buds regeneration of hybrid 717 was MS+0.5 mg/L 6 BA+0.2 mg/L NAA;The optimum medium for adventitious buds regeneration of hybrid 353 was MS+0.5 mg/L 6-BA+0.2 mg/L NAA+0.01 mg/L TDZ;The optimum medium for rooting of shoots was 1/2 MS+0.2 mg/L IBA+0.2 mg/L NAA, with a rooting efficiency of 100%. The best leaves for adventitious buds regeneration were on the tip 1st~ 3rd leaf of stems. 20 mg/L kanamycim could inhibit the bud regeneration of the hybrid 717 and 353,40 mg/ L kanamycim could inhibit the root regeneration of the hybrid 717 and 353 from the shoots.
