摘要
目的研究β-淀粉样肽(β-amyloid,Aβ)代谢改变与急性缺血性卒中(acute ischemic stroke,AIS)的关系。方法选取40例急性缺血性卒中患者和40例年龄相当的正常人作为对照,应用双抗体夹心酶联免疫吸附法(ELISA),测定了血液中可溶性Aβ1-40及Aβ1-42的水平。结果AIS患者血清Aβ1-40水平较正常对照显著升高(P<0.01),而血浆Aβ1-42水平较正常对照显著降低(P<0.01),血液中Aβ1-42/Aβ1-40及总体Aβ水平较正常对照显著降低(P<0.01)。结论急性缺血性卒中过程中存在的β淀粉样肽蛋白(β-amyloid precursor protein,APP)表达上调以及相伴随的短暂Aβ水平变化,可能是脑急性损伤和灌流不足的结果。
Objective To studay the relationship between the metabolic alternation of β-amyloid(Aβ) and acute ischemic stroke (AIS). Methods We prospectively enrolled 40 patients with acute ischemic stroke and 40 age-matched controls to detect the blood Aβ levels,which were determined with the double sandwich enzyme linked immunosorbent assays (ELISA). Results We found that patients with AIS showed markedly increased Aβ1-40 levels ( P 〈 0. 01 ), significantly decreased Aβ1-42 levels ( P 〈 0. 01 ), significantly decreased Aβ1-42/Aβ1-40 and Aβ in the blood compared with non-ischemic eontrols(P 〈0. 01 ). Conclusion Our data suggest that during the course of acute ischemic stroke, the expression of β-amyloid precursor protein(APP) is up-regulated and associated with transient diversity of Aβ levels, which might be derived from brain as a consequence of acute ischemic insults and hypoperfusion.
