复方糖耐康对GK大鼠胰脏的组织学影响 被引量:2

摘要 目的:观察中药复方糖耐康对自发性2型糖尿病GK大鼠胰脏组织病理变化的影响。方法:将50只SPF级GK大鼠按照血糖值随机分为5组:模型组、吡格列酮组、糖耐康低剂量组、糖耐康中剂量组、糖耐康高剂量组,并设Wistar大鼠10只作为正常对照组,连续给药8周,测定OGTT、空腹血清胰岛素、并计算胰岛素抵抗指数、观察胰脏的病理变化。结果:与正常对照组比较,GK大鼠空腹血糖正常或轻度升高,负荷2h后血糖明显升高;空腹血清胰岛素水平升高,呈高胰岛素抵抗状态;镜下观察到胰岛形态不规则,胰岛数及岛内细胞数目减少,胰岛细胞颗粒脱失,核大小不等,胰岛细胞被增生血管纤维所分隔,胰岛纤维化。糖耐康各剂量组及西药吡格列酮组对血糖、血清胰岛素及胰岛素抵抗指数以及胰腺病理变化均有不同程度改善,与模型组比较有显著差异。结论:中药复方糖耐康可通过降低血糖,改善胰岛素抵抗而发挥对胰岛细胞的保护作用。 Objective : To observe the effect of Tang - Nai - Kang on pathological changes of pancreas in spontaneous diabetic GK rat. Method:50 GK rats in SPF grade were divided into 5 groups according to plasma glucose level randomly, which were model group, Pioglitazone group, TNK - low - dosage group, TNK - middle - dosage group and TNK - high - dosage group. And 10 Wistar rats were as normal control group. After 8 weeks' treatment, oral glucose tolerance test (OGTT) ,fasting insulin level (Fins) and calculating insulin resistance index (HOMA - IR) were tested, and pathological changes of pancreas were observed. Result: Contrasted with normal control group,GK rat's fasting plasma glucose (FPG) was normal or increased mildly,2 hours postload glucose,Fins and HOMA - IR obviously increased. The morphous of pancreaticislets was irregular and the number of islets of pancreas or islet cell decreased. The particles of islet cells were also decreased, the sizes of nuclear were inequal. Islet cells were divided by hyperplastic vascellar fiber and pancreatic islets grew into fibrosis. Every TNK group and Pioglitazone group markedly improved plasma glucose, Fins, HOMA - IR and pathological changes of pancreas, there were significant difference comparing with model group. Conclusion:Tang -Nai -Kang can protected islet cell by decreasing plasma glucose and improving insulin resistance.
