摘要
[目的]研究以95%甲醇和碱液做溶剂从橘皮中提取橙皮苷的工艺条件。[方法]通过对溶剂、甲醇浓度、温度、pH值、浸提次数进行试验确定提取橙皮苷的最佳工艺条件。[结果]以桔皮为原料,用碱液和甲醇做溶剂提取橙皮苷的最佳工艺条件为:粗苷∶甲醇∶碱液[W(NaOH)为30%]=113∶0∶8,在85℃以上浸泡4 h,调节pH值为4。即先将甲醇和碱液搅拌混合,再将粗苷加入,继续搅拌1 h使粗苷溶解完全,然后过滤至滤液澄清,用盐酸调滤液的pH值至4,静置沉淀,过滤,滤饼用煮沸过的水洗涤至无色,甲醇滤液精馏回收,橙皮苷的收率达65%以上。[结论]此方法操作简便,易控制且提取橙皮苷的效率较高。
[ Objective ] The purpose was to research the technological condition of extracting hesperidin from orange peel with 95% methanol and alkali liquor as solvent. [ Method ] Through the experiment conducted on solvent, methanol eonen., temperature, pH value and extraction times, the optimum technological condition of extracting hesperidin was determined. [ Result ] With orange peel as raw material, the optimum technological condition of extracting hesperidin with alkali liquor and methanol as solvent was as follows: crude hesperidin: methanol: alkali liquor [ W(NaOH) = 30% ] = 11:30:8, soaking for 4 h at the temperature over 85 ℃ and adjusting pH value to be 4. Concretely, mixing and stirring methanol and alkali liquor and then adding crude hesperidin into the solution and going on stirring for 1 h till the crude hesperidin was dissolved completely, filtrating solution of crude hesperidin till the filtrate became clear, adjusting pH value of filtrate to be 4 by using hydrochloride, standing for precipitation and filtrating, washing filtrated cake to be colorless with boiled water, recovering methanol filtrate by distillation. Under this technological condition, the yield of hesperldin was higher than 65%. [ Conclusion ] This method was convenient to operate, easy to control and its efficiency of extracting hesperidin was relatively high.
