牛乳中嗜冷菌分离培养基筛选及其优化的研究 被引量:1

摘要 采用平板涂抹分离法研究培养基种类、增殖因子对牛乳中嗜冷菌分离效果的影响。结果表明:筛选嗜冷菌的最佳培养基为改良LA。针对嗜冷菌的营养需要,采用正交实验优化基础培养基及增殖因子的用量,得出最佳基础培养基配比:胰蛋白胨1.5%,葡萄糖1.0%,乳糖1.25%,酵母膏1.0%(均为质量分数);最佳增殖因子添加量为NaCl 0.5%。最后使用优化出的增菌培养基测定生长曲线,同时测定OD值的变化确定增菌培养终止时间为36 h,通过平板菌落计数,此时菌数可达9.3×105 cfu/mL。 The influence of medium variety, growth promoting factors to the isolation effect of Psychrophiles was studied by plate paint isolation methods. According to the nutritional needs of Psychrophiles basis culture medium and growth promoting factors were optimized by quadratic design. The optimum basis culture medium composition was as following: 1.5% peptone, 1.0% glucose, 1.25% lactose, 1.0% yeast extract. The optimum growth promoting factors were as following: NaCl 0.5%. Growth curve with optimized stimulated culture were determined by following the change of pH and OD. According to the growth curve, the terminal time was at 36 hours, Psychrophiles got to 9.3×10^5 cfu/mL in this culture for 36 hours at 7 ℃.
