双氢青蒿素对大鼠照射后早期肺组织病理形态学的影响 被引量:6

摘要 目的探索双氢青蒿素对大鼠放射性肺损伤的防治作用。方法采用γ射线单次全肺照射15 Gy,建立SD大鼠放射性肺损伤模型。108只同体重雄性SD大鼠,随机分为3组(n=36)。正常对照组灌服等体积溶媒;单纯照射组灌服等体积溶媒+全肺单次照射15 Gy;照射用药组灌服双氢青蒿素+全肺单次照射15 Gy。双氢青蒿素和溶媒皆于照射前3天开始灌服,持续至照射后15天。双氢青蒿素灌服剂量为60 mg.kg-1.d-1。对各组大鼠的肺外观、肺组织中炎症渗出、胶原纤维生成状况进行观察和比较。结果照射用药组大鼠肺较单纯照射组肺表面光滑、弹性好、充血水肿表现轻。单纯照射组大鼠肺组织炎症渗出及纤维化征象最严重。结论双氢青蒿素能够明显改善大鼠放射性损伤肺组织的炎性渗出,抑制胶原纤维的产生,减少血管内皮细胞和肺泡上皮细胞损伤,对放射性肺损伤具有一定的防治作用。 Objective To investigate the effect of dihydroartemisinin on radiation-induced histopathological changes in the lungs in the animal model of rats.Methods 108 SD rats were randomly divided into 3 groups(n=36 each): normal control group(group C,drenching of solvent alone),radiation control group(group R,drenching of solvent + single radiation of 15 Gy),treatment group with dihydroartemisinin and radiation(group DR).The group R and DR were treated at the dose of 60 mg·kg^-1·d^-1).The group R and DR were radiated thorax with ^60Coγ at the dose of 15 Gy/1 Fraction after chloralization.Drenching once a day began on the 3rd day before radiation until the 15th day after radiation.Changes of lung surface and histopathological were observed.Results After radiation,the swelling appearance,flexibility and hemorrhage of the lung surface of group DR were better than group R.By H-E and VG staining,the inflammatory effusion and fibrosis in group R were the severest,group DR was between the group C and R.By transmission electron miscroscope,the injury of type Ⅱ pneumonocytes and endothelial cells of capillary in R is worse than in DR.Conclusion Dihydroartemisinin may relieve inflammation and fibrosis in the radiation-induced pulmonary injury of rats,thus contributing to the prevention and treatment of radiation-induced pulmonary injury.
