墨兰的无菌播种及根状茎的增殖研究 被引量:15

摘要 [目的]研究墨兰无菌播种和根状茎的增殖情况,为墨兰的组织快繁提供参考。[方法]对金嘴墨兰的种子进行无菌播种,并对不同的培养基成分与培养条件在墨兰根状茎生长、增殖和生根的影响进行了研究。[结果]金嘴墨兰较适宜的种子萌发培养基为1/2 MS或改良MS;在根状茎增殖过程中,KC培养基的增殖效果最好,增殖倍数为8.1,其次为MS培养基,增殖倍数为7.4;6-BA和NAA的比例控制在2∶1效果较好;在墨兰生根培养过程中所加入的5种营养附加物香蕉泥、椰子汁、红薯汁、马铃薯汁、玉米汁都能够促进生根。[结论]根状茎增殖的较适宜培养基为KC+6-BA 2.0 mg/L+NAA 1.0 mg/L+蔗糖30 g/L+琼脂7.1 g/L+0.1%炭粉;红薯汁对墨兰的生根有显著的促进作用。 [Objective] The research aimed to study the aseptic sowing and rhizome proliferation of Cymbidium sinense, and provide references for rapid propagation of aseptic sowing. [ Method] The article studied the aseptic seedling, growth, rhizome proliferation and rooting of jinzui Cymbidium sinense. [ Result] The results showed that 1/2 MS and the improved MS medium were suitable for seed germination. Banana slurry, coconut milk, batata milk, potato juice, corn juice could promote rooting. In the course of rhizome proliferation, KC medium was the best, which proliferation times was 8.1, MS medium was better which proliferation times was 7.4. The propotion 2:1 of 6-BA and NAA obtained better effect. [Conclusion] The suitable medium for proliferation of rhizome was KC+6-BA 2. 0 mg/L + NAA 1.0 mg/L + Sugar 30g/L + near 7.1g/L+carbon powder 0.1%.Batata milk was more suitable for root growth of jinzui Cymbidium sinense.
