摘要
目的:观察鳖甲不同剂型防治大鼠肝纤维化效果并探讨其作用机制。方法:用四氯化碳(CCl4)诱导形成大鼠肝纤维化模型,采用鳖甲不同剂型进行预防和治疗,并与正常对照组、模型对照组相比较,观察各组大鼠病理组织学、肝组织胶原含量、免疫组化细胞因子的表达、血清透明质酸、肝功能、肝组织氧化指标,透射电镜观察肝组织超微结构的改变等。结果:鳖甲煎煮液组SSS计分、肝组织胶原含量、纤维化指标、促肝纤维化因子、血清酶学指标显著低于模型组与鳖甲微粉组,而抗氧化指标显著上升(P<0.05~0.01)。结论:鳖甲煎煮液能有效地预防和治疗大鼠肝纤维化,效果显著,其作用机制可能是通过抗脂质过氧化、改善肝组织病理、改善肝功能、调控细胞因子水平等而发挥抑制肝星状细胞(HSC)活化增殖及细胞外基质(ECM)合成分泌、促进ECM降解吸收等综合作用,阻断和治疗肝纤维化。鳖甲微粉药液无此作用。
Objective : To evaluate the effects of different dosage forms of Amydae Carapax on prevention & treatment of liver fibrosis in rats, and to explore their action mechanism. Methods: Liver fibrosis was successfully induced in 100 rats by subcutaneous injection of carbon tetrachloride( CC14 ) ,then some rats were prevented and treated with amydae carapax decoction or amydae carapax micro - powder. At the same time, positive nad normal control were used. At the end of animal experiment, semi - quntitative histopathological scores of inflammation and fibrosis, content of collagen of the liver, levels of liver some cytokines, serum hyaluronic acid, liver function and liver tissue oxidation index were compared, the nhrastructnre of liver biopsy was observed with transmission electricity. Results : Compared with those in model group and prevention & treatment group of amydae carapax micro- powder, histopathological scores of fibrosis, collagen content in the liver, liver fibrosis index and some indexs in serum were significantly lower, but liver tissue anti - oxidation index was significantly higher ( P 〈 0.05 - 0.01 ) in prevention & treatment group of amydae carapax decoction. Conclusion: Treatment with amydae carapax decoction could resist lipid perioxidation, improve pathological function of liver tissue, improve liver function and regulate cell factor level. Thus it could inhibit hepatic stellate ceils (HSC) activated multiplication and extra cellular matrix (ECM) synthesis and secretion, promote ECM degradation and significantly prevent and treat liver fibrosis in rats. But amydae carapax micro - powder did not prevent and treat liver fibrosis.
