己酮可可碱对内毒素血症大鼠心肌HSP70和NF-κB的影响 被引量:2

Effects of Pentoxifylline on the Expression of HSP70 and NF-κB in Myocardial Tissue in Rats with Endotoxemia

摘要 目的观察己酮可可碱(pentoxifylline,PTX)对内毒素血症大鼠心肌热休克蛋白70(HSP70)和核因子-κB(NF-κB)的影响,探讨PTX对内毒素血症大鼠心肌损伤的保护作用及其机制。方法采用直接注射内毒素的方法建立大鼠急性内毒素血症模型。18只Wistar大鼠随机分成3组:假手术组、内毒素组、PTX组。采用Western blot检测心肌胞质蛋白内HSP70、抑制蛋白-κB(I-κB)和核蛋白内NF-κB的表达,同时观察心肌病理形态。结果内毒素组心肌组织内HSP70和NF-κB蛋白水平均明显增高,而I-κB蛋白水平明显降低,与假手术组比较差异有极显著性意义(P<0.01);予PTX干预后,心肌组织HSP70蛋白水平进一步增高,而NF-κB蛋白水平却明显下降,与内毒素组比较差异有极显著性意义(P<0.01)。结论PTX可以通过增加内毒素血症大鼠心肌组织内HSP70蛋白表达和降低NF-κB活性,对心肌产生保护作用。 Objective To study the effects of pentoxifylline (PTX) on the expression of HSP70 and NF-κB in myocardial tissue in rats with endotoxemia and explore the protective effects of PTX on the myocardial injury induced by endotoxemia and the mechanism. Methods The models of acute endotoxemia in rats were established by the method of injecting lipopolysaccharide (LPS) directly. Eighteen Wistar rats were randomly divided into 3 groups: sham-operated group (S), LPS group (L) and PTX-treated group (P). Western blotting was used to detect the expression of HSP70 and NF-κB in the myocardial tissues. The pathologic lesions of myocardial tissues were observed. Results The levels of HSP70 and NF-κB proteins in the myocardial tissues in L group were higher, but the levels of I-κB protein were lower than those in S group (both P〈0.01). After treatment with PTX, the levels of HSP70 protein in the myocardial tissues in P group were significantly increased, but the levels of NF-κB were significantly reduced as compared with L group (both P〈0.01). Conclusion PTX can protect the myocardial tissues by elevating the expression of HSP70 and reducing the expression of NF-κB in the myocardial tissues of rats with endotoxemia.
